响应面优化UAE积雪草中积雪草苷工艺 被引量:2

The Optimized Asiaticoside Extraction Process by Response Surface UAE Methodology from Centella asiatica

摘要 为优化提取积雪草中积雪草苷工艺试验条件,利用单因素变量法考察单变量因素,响应面优化工艺参数,寻求最佳提取条件,启用高相液相色谱仪测其积雪草苷含量。结果表明,在最优工艺条件(温度35℃,提取时间2.5h,固液比1︰35 g/mL,超声波功率240 W)下,相对标准偏差为0.39%, RSD值小于2%,仪器检出限为0.000 000 041 02μg/m L。试验样品a为低含量0.1 mg/mL标准液:积雪草苷的回收率在100.2%~101.0%之间, RSD为0.03%;样品b为高含量1.0 mg/mL标准液:积雪草苷的回收率在100.1%~100.2%之间, RSD为0.01%;积雪草苷含量达6.4 mg/g,满足效能指标的要求。该方法是一项新的试验尝试,试验结果令人满意。 The extraction process of Asiaticoside from Centella asiatica experimental condition was optimized. In order to seek the best extraction condition, the single factor variable method and optimization of process parameters by response surface methodology were determined, using maximum absorption wavelength spectrophotometric determination of asiaticoside standard. The result showed that the optimum condition was the temperature of 35 ℃, the extraction time of 2.5 h, the ratio of solid to liquid of 1︰35 g/mL, and the ultrasonic power of 240 W. The relative standard deviation was 0.39%, the RSD value was less than 2%, and detection limit was 0.000 000 041 02 μg/mL. The experimental sample a was a low content 0.1 mg/mL standard solution: The recovery rate of asiaticoside was between 100.2% and 101.0%, and RSD was 0.03%. The experimental sample b was a high content 0.1 mg/mL standard solution: the recovery rate of asiaticoside was between 100.1% and 100.2%, and RSD was 0.01%. The content of asiatiside had reached 6.4 mg/g, thus it could meet the requirements of performance indicators. This method was a new experiment with satisfactory results.
