摘要
目的观察黄芩汤对结肠炎相关结肠癌小鼠模型Notch/Wnt信号通路的影响。方法 C57BL/6雄性小鼠随机分为正常组、模型组、低剂量和高剂量黄芩汤组,用氧化偶氮甲烷联合葡聚糖硫酸钠建立结肠炎癌变动物模型。观察疾病活动指数(DAI)、结肠长度和肠道肿瘤数量,ELISA检测血清促炎因子TNF-α、IL-6和IL-1β含量,免疫组织化学检测结肠上皮细胞增殖、凋亡及Notch/Wnt信号通路中关键蛋白表达。结果黄芩汤显著降低模型小鼠DAI评分和肿瘤数量,增加结肠长度,抑制结肠上皮细胞增殖并促进凋亡,下调促炎因子(TNF-α、IL-6、IL-1β)、Notch-1、Hes-1、β-catenin、c-myc表达。结论黄芩汤可有效预防结肠炎相关结肠癌的发生与发展,其机制可能涉及Notch/Wnt信号通路的抑制。
Objective To investigate the effect of Huangqin Tang(HQT) on Notch/Wnt pathway in mice with colitisassociated colon cancer(CACC). C57 BL/6 male mice were randomized to normal, model, and low-and high-dose MethodsHQT groups. CACC models were established using azoxymethane and dextran sulfate sodium. The disease activity index(DAI), colon length and number of tumors were observed. Serum levels of proinflammatory cytokines(TNF-α, IL-6 and IL-1β), proliferation, apoptosis, and key protein expression of Notch/Wnt pathway were determined by ELISA and immunohistochemistry. Results HQT significantly decreased DAI and tumor number but increased colon length. Furthermore, HQT dramatically suppressed proliferation but enhanced apoptosis, and down-regulated expression of proinflammatory cytokines(TNF-α, IL-6 and IL-1β), Notch-1, Hes-1, β-catenin, and c-myc. Conclusion HQT may prevent the development and progression of CACC via inhibiting Notch/Wnt signaling.
