摘要
目的观察功能未知基因C6orf120敲除后对大鼠表型变化的影响,初步探究该基因的功能。方法将雄性野生型(wild type,WT)大鼠作为对照组,C6orf120基因敲除(C6orf120-/-)大鼠作为实验组,称量子代大鼠20 d体质量及观察子代生长情况。利用逆转录聚合酶链式反应(reverse transcription PCR,RT-PCR)技术检测大鼠肝组织TNF-α的转录水平。利用Western blotting技术分析6~8周WT大鼠和C6orf120-/-大鼠肝脏组织TNF-α和Cleaved caspase-3的表达情况。结果与WT大鼠相比,C6orf120-/-子代大鼠出生20 d体质量显著降低(t=5. 292,P <0. 0001)。成年C6orf120-/-大鼠TNF-α、Cleaved caspase-3在肝组织中的表达水平显著高于WT大鼠(P=0. 0172、0. 0301)。结论基因C6orf120可能对大鼠肝功能具有一定的保护作用,其保护机制可能与抗炎和抗凋亡有关。
Objective To observe the effect of the knock-out of unknown function gene C6 orf120 on the phenotypic changes in rats and investigate the preliminary function. Methods The rats were divided into control group（ wild type,WT） and experimental group（ C6 orf120^-/-）. The weight of rats at 20 days were recorded. The expression levels of inflammatory protein TNF-α and apoptosis protein Cleaved caspase-3 in 6-8 weeks of WT and C6 orf120-/-rats liver tissue were analyzed by RT-PCR and Western blotting. Results Compared with WT rats,C6 orf120^-/-rats were significantly reduced in body weight at 20 days（ t = 5. 292,P 0. 0001）. TNF-α,Cleaved caspase-3 expression levels of adult C6 orf120^-/-rats in liver tissue were significantly higher than those in WT rats（ P = 0. 0172,0. 0301）.Conclusion Gene C6 orf120 may have a protective effect on rat liver function,and its protective mechanism may be related to anti-inflammatory and anti-apoptosis.
