摘要
目的:采用HPLC法测定大鼠血浆中替格瑞洛的浓度。方法:血浆样品采用甲醇沉淀蛋白,色谱柱为Inertsil ODS-3色谱柱(150 mm×4.6 mm,5μm),流动相为乙腈-10 mM乙酸铵水溶液(50∶50),流速为1.0 ml/min,紫外检测波长为299 nm。结果:大鼠血浆中替格瑞洛在0.1~5μg/ml范围内线性关系良好,标准曲线为A=32 465C+935.38,相关系数r=0.999 1。替格瑞洛的低、中、高浓度的提取回收率分别为85.21%、86.91%和85.87%;日内、日间精密度RSD均小于10%。结论:本法简便、快速、可靠,适用于检测血浆中替格瑞洛的浓度。
Objective:To establish an HPLC method for determination of Ticagrelor in rat plasma.Method:The rat plasma samples were prepared by protein precipitation with methanol.HPLC analysis of plasma samples was performed with a Inertsil ODS-3(150 mm×4.6 mm,5 μm)column.The mobile phase consisted of a mixture of 10 mM ammonium acetate buffer and acetonitrile(50∶50,v/v),at a flow rate of 1.0 ml/min.The wavelength of 299 nm was used for detection.Result:A good linearity of ticagrelor in the rat plasma was shown in the range of 0.1-5 μg/ml(r=0.999 1).The regression equation was A= 32 465C+935.38.The extraction recoveries of ticagrelor in low,middle and high concentrations were 85.21%,86.91% and 85.87%,respectively.The intra-day and inter-day variation(RSD) were both less than 10%.Conclusion:The method is simple,fast,and reliable,and it can be employed to determine the concentration of Ticagrelor in plasma.
