自噬相关基因在肿瘤发生中的功能及作用机制 被引量:4

摘要 自噬是一种广泛存在于真核细胞中与溶酶体降解细胞组分相关的、进化上高度保守的、多种基因和信号通路共同调控的重要生理、生化过程。生物体依靠自噬维持细胞的自我保护、蛋白代谢和细胞内外环境平衡。近年来,随着对肿瘤研究的不断深入,发现肿瘤的发生、发展与自噬的紊乱密切相关。多种自噬相关基因(ATGs)通过不同的信号调节通路和机制,对肿瘤进行双重调节。本文着重介绍自噬在肿瘤发生、发展中的作用,特别是几种重要的自噬相关基因在肿瘤发展中的功能及分子信号通路。 Autophagy is an important physiological and biochemical process widely existing in eukaryotic cells, which is associated with lysosomal degradation of cell components and highly conserved in evolution and regulated by a variety of genes and signaling pathways. Organisms rely on autophagy to maintain cell self-protection, protein metabolism and the balance of intracellular and extracellular environment. In recent years, with the deepening of cancer research, it is found that the occurrence and development of tumors are closely related to the disorder of autophagy. Multiple autophagy-related genes(ATGs) conduct double-regulation for tumors through different signaling pathways and mechanisms. This review focuses on the role of autophagy in tumorigenesis and development, especially the functions and molecular signaling pathways of several important autophagy-related genes in tumorigenesis.
