摘要
目的:系统研究蓝花参的生药学特征,为其品种鉴别、质量标准制定及进一步开发利用提供参考。方法:基于数码显微镜与电子目镜组合的Digital-Camera 6.0软件环境下,采用微性状观察法对蓝花参全株进行表面性状观察;采用冷冻切片技术制备组织切片,并进行显微观察、拍照、测量,记录相关数据;利用Adobe Bridge CS6和Photoshop CS6软件对图片进行景深叠加处理。结果:根以密集的皮层和韧皮部乳汁管群、菊糖晶体为主要鉴别点;茎以表面的平轴状不定式气孔、棱脊处的厚角组织、表面的单细胞非腺毛、内皮层细胞、较多的单个分布的韧皮部乳汁管、纤维束为主要鉴别点;叶以具角质层的近方形表皮细胞、不定式气孔、单细胞非腺毛、外韧型维管束、明显的内皮层环为主要鉴别点;以花瓣边缘明显的乳状凸起、花粉粒形态、果实和种子形态大小、种子表面的皱脊和条状纹理等为其余鉴别点;蓝花参粉末以众多的多纹孔纤维、平轴状不定式气孔、单细胞型非腺毛和不规则形状菊糖、不规则浅波齿状叶表皮细胞、具纵向条纹饰的长椭圆形种子、散在的无色透明油滴和具3个萌发孔的圆形花粉粒为主要鉴别点。结论:根、茎、叶的横切面特征以及花、果实、种子的表面特征是蓝花参生药的主要鉴别点。
Objective: To systematically study on the pharmacognosy characteristics of Wahlenbergiae Marginatae Radix seu Herba,in order to provide references for its identification,quality standards and further development and utilization. Method: Under Digital-Camera 6. 0 software environment combined with digital microscope and electronic eyepiece,microscopic character observation was used to observe surface characteristics of the whole plant. Tissue sections were prepared by frozen section method, then microscopic observations,photographs,measurements,and related data were recorded. The superposition of depth of focus was treated by Adobe Bridge CS6 and Photoshop CS6 softwares. Result: Roots of Wahlenbergiae Marginatae Radix seu Herba had lots of laticifer groups and irregular-crystal inulins in cortex and phloem; its stems had many infinitive stomata in flat axis, collenchymas, single-cell non-glandular hairs, endodermis, single distributed laticifers and fiber bundles. The main identificational points in its leaves were squarish epidermal cells with cuticle, infinitive stomata,single-cell non-glandular hairs,collateral vascular bundle and obvious endothelium ring. And also there were lots of other points in the crude herb,such as protuberances in the petals margins,pollen morphology,the size of fruits and seeds, wrinkles and longitudinal stripes in the surface of seeds. Hence, the microscope characteristics of the powder of Wahlenbergiae Marginatae Radix seu Herba contained lots of multi-hole fibers,infinitive stomata in flat axis,single-cell type non-glandular hairs,inulins in irregular crystals; and also there were leaf epidermal cells of shallow waves,oblong seeds of longitudinal stripes,scattered colorless transparent oil drops,round pollen grains of three germinating holes in the medicinal powder; the above aspects could be used as the main identificational points of its powder.
