摘要
目的:研究柴胡作为引经药与大黄-丹参药对配伍对大鼠肝纤维化的影响。方法:通过腹腔给予CCl_4复制SD大鼠肝纤维化的模型,实验设空白组、模型组、大黄-丹参药对组(5.0 g·kg^(-1))、药对+柴胡低、中、高剂量组(5.0 g·kg^(-1)+0.75 g·kg^(-1)、5.0 g·kg^(-1)+1.5 g·kg^(-1)、5.0 g·kg^(-1)+3.0 g·kg^(-1))。从第3周开始,各组大鼠按相应剂量灌胃给药6周,观察大鼠肝组织形态的病理改变,检测血中ALP、ALT、AST的活性,ELISA法测定Ⅲ型前胶原(PCⅢ)、透明质酸(HA)和层黏连蛋白(LN)的含量,酸水解法分析肝中羟脯胺酸(HYP)的含量,免疫组化法测定肝中转化生长因子-β1(TGF-β1)的表达。结果:与模型组比较,各给药组大鼠肝脏组织形态明显改善,血清中ALP、ALT、AST活性和PCⅢ、HA、LN的含量及肝中TGF-β1的表达降低,药对+柴胡组降低更明显。结论:大黄-丹参药对能明显改善SD大鼠的肝纤维化程度,柴胡作为引经药可增强其疗效。
Objective： To observe the therapeutic effect of herb pair of Rhei Radix et Rhizoma and Salviae Miltiorrhizae Radix et Rhizoma with Bupleuri Radix guides in hepatic fibrosis rats. Methods： The hepatic fibrosis rat models were induced by intraperitoneal injection of CCl_4. Groups were divided into normal group, model group, herb pair group（5.0 g·kg^（-1））, herb pair plus low dose of Bupleuri Radix（5.0 g·kg^（-1）＋0.75 g·kg^（-1））, herb pair plus medium dose of Bupleuri Radix group（5.0 g·kg^（-1）＋1.5 g·kg^（-1）） and herb pair plus high dose of Bupleuri Radix group（5.0 g·kg^（-1）＋3.0 g·kg^（-1））. Then diferent drugs were administrated via gavage for 6 weeks from week 3 after modeling. Rats were sacrificed after the last administration, then their serum was collected to measure the activities of serum alanin aminotransferase（ALT）, aspartate aminotransferase（AST） and alkaline phosphatase（ALP）. Levels of laminin（LN）, procollagen Ⅲ（PCⅢ） and hyaluronic acid（HA） in serum were detected by enzyme linked immunosorbent assay（ELISA）. The content of hydroxyproline（HYP） in liver homogenate was measured by acid hydrolysis method. The expression of transforming growth factor β1（TGF-β1） in liver was analysed by immunohistochemical method. Results： Compared with the model group, the levels of ALT, AST, ALP, PCⅢ, HA, LN and the content of HYP in experimental groups were significantly decreased, the expression of TGF-β1 in liver was reduced, the liver pathological fibrosis was obviously relieve. Compared with the herb pair group, the levels of ALT, AST, ALP, PCⅢ, HA, LN and the expression of TGF-β1 in herb pair plus Bupleuri Radix group were significantly decreased. Conclusion： The herb pair of Rhei Radix et Rhizoma and Salviae Miltiorrhizae Radix et Rhizoma has protective effect on liver fibrosis rat, Bupleuri Radix could further improve the curative effect.
