单次运动对缺氧诱发心肌自噬反应的影响

Effects of Single Exercise on Myocardial Autophagy Induced by Hypoxia

摘要 为研究运动后不同休息时间对间歇性低氧诱发心肌自噬反应的影响,本研究将8周龄Sprague-Dawley雄性大鼠,随机分成控制组(control group,CON)、运动组(exercise group,EXE)、间歇性低氧曝露组(intermittent hypoxia group,IH)、空气曝露组(room air group,RA)、运动后1h(post-exercise 1 h,PE1h)和3h(PE3h)合并RA或IH组。大鼠给予单次持续8hIH(氧气浓度下降至2%~6%约1~2s/75s),单次运动方式以速度24m/min、坡度2%的强度在动物跑台上跑步60min。动物牺牲后,检测左心室心肌糖原、自噬反应相关蛋白及腺粒体功能相关mRNA表达量。研究发现,运动后心肌细胞微管相关蛋白1轻链3-Ⅱ(microtubule-associated protein 1 light chain 3-Ⅱ,LC3-Ⅱ)/LC3-Ⅰ表达量、糖原含量下降(p〈0.05),p62表达量没有变化(p〉0.05)。IH对心肌p62表达量没有影响(p〉0.05),与RA比较,IH的LC3-Ⅱ/LC3-Ⅰ、核内呼吸因子1和2(nuclear respiratory factor 1 and 2,NRF-1和NRF-2)mRNA表达量上升(p〈0.05)。与IH比较,PE1h+RA、PE3h+RA及PE3h+IH各组LC3-Ⅱ/LC3-Ⅰ、NRF-1和NRF-2mRNA表达量均下降(p〈0.05),虽然PE1h+IH的NRF-1和NRF-2 mRNA表达量下降(p〉0.05),但LC3-Ⅱ/LC3-Ⅰ表达量没有改变(p〉0.05)。本研究说明,IH曝露前3h进行运动,可避免IH造成心肌LC3-Ⅱ/LC3-Ⅰ表达量上升的现象。 To investigate the effects of different time intervals after exercise on intermittent hypoxia (IH)-induced autophagy in rat left ventricle, eight-week-old male Sprague-Dawley rats were randomly assigned to control (CON), exercise (EXE), IH, room air exposure (RA), post-exercise 1 h and 3 h combined with RA (PElh +RA and PE3h+RA) orIH (PElh+IHand PE3h+IH) groups. Rats were exposed to a single IHbout (2%-6% 02 for 1-2 s/75 s) for 8 h and a single exercise bout on treadmill (24 m/min, 2% grade) for 60 min. Myocardial glycogen, autophagy- related proteins and mitochondrial-related mRNAs were determined after euthanisation. It could be found that immediately after exercise, microtubule-associated protein 1 light chain 3- Ⅱ (LC3-Ⅰ/)/LC3-Ⅰ protein levels decreased (/9〈0.05) but p62 levels did not alter (p〉0.05). As compared with RA, IH did not alter p62 levels but did increase LC3- U/LC3-Ⅰ protein and nuclear respiratory factor 1 and 2 (NRF-1 and NRF-2) mRNA levels (p〈0.05). Compared with IH, significantly lower LC3-Ⅱ/LC3-Ⅰ protein and NRF- 1 and NRF2 mRNA levels were found in PElh+RA, PE3h+RA and PE3h+IH (p〈0.05). NRF-1 and NRF-2 mRNA levels were lower in PE 1 h+IH than inlH (p〈0.05), although LC- Ⅱ/LC3-Ⅰ levels were similar (p〉0.05). It could be concluded that exercise at 3 h before IH exposure could prevent IH-induced increased myocardial LC3-Ⅱ/LC3-Ⅰ levels.
