摘要
目的分析PAI-1(4G/5G)和ABCB1 C3435T基因多态性与狼疮继发股骨头坏死(SLE-ONFH)的相关性。方法纳入我院2016-06~2017-06符合入组标准的40例SLE患者的临床资料,其中SLE-ONFH组19例,无股骨头坏死(ONFH)的SLE患者21例为对照组。对受试者抽取空腹肘静脉血,用数字荧光分子杂交技术(DFMH)测定各组PAI-1(4G/5G)和ABCB1 C3435T基因多态性,其中PAI-1(4G/5G)基因4G4G型、ABCB1 C3435T基因CC型为高危型。结果 SLE-ONFH组和对照组PAI-1(4G/5G)基因4G4G型的频率分别为31. 6%,14. 3%,差异无统计学意义(χ~2=1. 71,P> 0. 05); SLE-ONFH组和对照组ABCB1 C3435T基因CC型的频率分别为52. 6%,19%,差异有统计学意义(χ~2=4. 94,P <0. 05)。结论 ABCB1C3435T基因与SLE-ONFH存在相关性,可能成为筛查SLE-ONFH高危人群的指标之一。
Objective To investigate the correlation between systemic lupus erythematosus (SLE)-related osteoneerosis of the femoral head(SLE-ONFH) and polymorphisms of PAL1 (4G/5G) gene and ABCB1 C3435T gene. Methods Forty SLE patients were enrolled with the 2012 SLICC Classification Criterion from June 2016 to June 2017, including 19 eases of SLE-ONFH( SLE-ONFH group) and 21 eases of non-SLE-ONFH( control group). The clinical ilfformation of 40 SLE patients was collected. The fasting blood of cubital vein was collected from each patient. The polymorphisnls of PAI-1 4G/5G gene and ABCB1 C3435T gene were detected by digital fluo- rescence molecular hybridization technique(DFMH). Results The frequency of PAI-1 4G/4G genotype was 31.6% in SLE-ONFH group and 14.3% in control group, but there was no significant difference(χ^2 = 1.71 ,P 〉 0. 05). The frequency of ABCB1 C3435T CC genotype in SLE-ONFH group was also higher than that in control group (χ^2 = 4. 94,P 〈 0. 05). Conlusion ABCB1 C3435T gene may be associated with the susceptibility to SLE-ONFH. ABCB1 C3435T CC genotype could be one of the sensitive hematological indexes for screening the high risk population of SLE-ONFH.
