摘要
目的观察环磷酰胺(CY)对严重烧伤大鼠早期心肌中性粒细胞浸润的影响,探讨CY保护心功能的作用机制。方法选取清洁级雄性SD大鼠100只,随机分为4组,CY组(n=40)、单纯烧伤组(n=40)、CY对照组(n=10)、空白组(n=10)。CY组与单纯烧伤组建立30%体表总面积Ⅲ度烧伤。CY组和烧伤组在建模成功后2h、6h、12h、24h时相点记录左心室收缩压(LVSP)、左心室终末舒张压(LVEDP)、左室压最大上升速率(dp/dtmax)、左室压最大下降速率(-dp/dtmax)。检测大鼠血清肌钙蛋白I(cTnI)及肌酸激酶同工酶(CK-MB)。取大鼠心脏组织锡纸包裹后立即置入-80℃冰箱保存用于检测髓过氧化物酶(MPO)。同法读取空白组及CY对照组各项参数及心肌取材。结果 CY对照组与空白组比较,LVSP、LVEDP、dp/dtmax、-dp/dtmax值,心肌MPO活性,血清cTnI和CK-MB含量比较,差异无统计学意义(P>0.05);CY组各时相点LVSP、LVEDP、dp/dtmax、-dp/dtmax值显著高于烧伤组(P <0.05);CY组各时相点MPO、血清cTnI和CK-MB含量均明显低于烧伤组(P <0.05)。结论低剂量CY能显著改善心肌收缩及舒张功能,抑制严重烧伤大鼠心肌组织MPO、cTnI及CK-MB的释放,具有明显的心肌保护作用。
Objective To observe the effect of cyclophosphamide on neutrophile granulocyte infiltrated in cardiac muscle in the earlystage of severely burned rats,To study cyclophosphamide's mechanism of action about protection of myocardial function.Methods One hundred clean class male rats(SD rats)were randomly divided into four groups:CY group ( n =40),burn group ( n =40),controlgroup ( n =10) and the normal group ( n =10).The model of 30% TBSA third degree burn was established in the CY group and theburn group.To record LVSP,LVEDP,dp/dtmax,and dp/dtmax at 2,6,12 and 24 hours after burn injury of CY group and burn group.Tro -ponin I(cTnI),and creatine kinase isoenzyme(CK MB) in serum of rats were detected.The rat heart was wrapped in tinfoil and put im -mediately in refrigerator at -80 ℃,for the test of myeloperoxidase(MPO).The same method was used in the normal group and controlgroup.Results Compared with normal group,the CY control group showed no obvious difference of LVSP,LVEDP,dp/dtmax, -dp/dt -max,MPO in cardiac muscle,cTnI and serum CK MB(P 〉0.05).LVSP,LVEDP,dp/dtmax,-dp/dtmax of the CY group were obviouslyhigher than those in burn group at each time point(P 〈0.05).MPO,cTnI and CK MB in serum of the CY group were obviously lowerthan those in the burn group at each time point(P 〈0.05).Conclusion Low dose CY could obviously improve systolic and diastolicfunction of heart;reduce the release of MPO,cTnI,and CK MB in cardiac muscle of severely burned rats;and protect the function ofthe cardiac muscle.
