摘要
以健康植株上的饱满芽和茎段为试材,采用组织培养方法,建立了李、杏砧木品种‘St.Julien A’无性繁殖技术体系,以期为李和杏规模化生产奠定基础。结果表明:以枝条上饱满的芽为外植体,以MS+BA 1.0 mg·L^(-1)+IBA 0.1mg·L^(-1)作为初代培养基、WPM+BA 1.0mg·L^(-1)+IBA 0.1mg·L^(-1)作为增殖培养基、1/2MS+IBA 1.0mg·L^(-1)作为生根培养基,李、杏砧木品种‘St.Julien A’增殖系数可达8.5,生根率达96%;在蛭石基质中,移栽成活率达到91.6%。
The full buds and shoot segments of healthy plants were used for in vitro culture,a complete and practical micropropagation protocol for rapid vegetative propagation of the rootstock‘St.Julien A'was developed.The results showed that using fully developed buds as explants,MS+BA 1.0 mg·L^-1+IBA0.1 mg·L^-1 as the initial medium,WPM+BA 1.0 mg·L^-1+IBA 0.1 mg·L^-1 as proliferation medium and 1/2 MS+IBA 1.0 mg·L^-1 as rooting medium,the proliferation coefficient and the rooting percentage reached 8.5 and 96%,respectively.Transplanted in the substrate of vermiculite,the survival percentage of transplanting plantlets reached 91.6%.
