摘要
为研究日本蛇根草功能基因的表达提供内参基因,笔者参照GenBank中桔梗的肌动蛋白基因(Actin)序列设计引物,以日本蛇根草的cDNA为模板,通过RT-PCR方法成功克隆获得日本蛇根草Actin基因片段,并将其命名为OjActin(OjActin片段长469 bp)。功能保守域分析显示OjActin归属于Actin超家族,与其他植物Actin的氨基酸序列同源性达94%以上;系统进化树分析显示,OjActin与咖啡的亲缘关系最近。
In order to provide control gene for the study of functional genes expression of Ophiorrhiza japonica , we designed the primers on the basis of Aticn from Eustoma in the Genebank. We successfully cloned the Actin gene fragment by RT-PCR method using the cDNA of Ophiorrhiza japonica as template, and named it as OjActin( OjActin was 469 bp and belonged to the Actin superfamily). OjActin shared over 94% amino acid sequence homology with Actins from other plants, and had the nearest relationship with Actin from Coffea canephora according to polygenetic analysis.
