摘要
目的观察益气补肾方对自然流产模型小鼠母-胎界面叉头翼状螺旋转录因子3(Foxp3)、维甲酸相关核孤儿受体γt(RORγt)、转化生长因子-β(TGF-β)、肿瘤坏死因子-α(TNF-α)蛋白表达及妊娠结局的影响。方法 CBA/J雌鼠与BALB/c雄鼠交配建立正常妊娠模型6只;CBA/J雌鼠与DBA/2雄鼠交配建立小鼠自然流产模型,孕鼠随机分为自然流产模型组、环孢素组、益气补肾高、中、低剂量组,每组6只。正常妊娠模型组和自然流产模型组给予生理盐水(10mL/kg);环孢素组给予环孢素A溶液(10mL/kg);益气补肾高、中、低剂量组分别给予12、24、48g/kg益气补肾方;均连续14天。给药结束后,处死孕鼠,观察子宫胚胎数量并计算胚胎吸收率;Western blot法检测母-胎界面Foxp3、RORγt蛋白表达,免疫组化法检测TGF-β、TNF-α蛋白表达。结果 (1)与自然流产模型组比较,环孢素组及益气补肾低、中、高剂量组胚胎吸收率均明显降低(11.9%、14.3%、11.1%、13.2%比25.0%,P均<0.05);益气补肾各剂量组胚胎吸收率与环孢素组相比,差异无统计学意义(P>0.05);益气补肾各剂量组间胚胎吸收率差异无统计学意义(P>0.05)。(2)与自然流产模型组比较,环孢素组、益气补肾中、高剂量组小鼠Foxp3蛋白表达显著升高[(1.223±0.240)、(1.220±0.241)、(1.383±0.216)比(0.700±0.147),P<0.05],益气补肾中、高剂量组与环孢素组间差异无统计学意义(P>0.05)。与自然流产模型组比较,环孢素组、益气补肾中、高剂量组RORγt蛋白表达显著降低[(0.950±0.246)、(0.897±0.321)、(0.670±0.082)比(1.456±0.165),P<0.05],益气补肾中、高剂量组与环孢素组间差异无统计学意义(P>0.05)。(3)与正常妊娠模型组比较,自然流产模型组胎盘TGF-β表达显著降低[(0.033±0.015)比(0.084±0.033),P<0.05]。与自然流产模型组比较,环孢素组及益气补肾方低、中、高剂量组TGF-β表达均显著升高[(0.102±0.046)、(0.112±0.030)、(
Objective To observe the effects of replenishing qi and tonifying kidney recipe on the expression of Foxp3（foxhead box P3）, RORγt（rar-ralated orphan receptor -gamma-t）, TGF-β（transforming growth factor-β）andTNF-α（tumor necrosis factor-α）molecules, and pregnancy outcome at the maternal-fetal interface of spontaneousabortion model mice, in order to reveal the mechanism of materno fetal immunoregulation and to provide provideda theoretical basis for the Prevention and cure of recurrent spontaneous abortion. Methods CBA∕J female mouseand BALB∕c male mouse to produce a normal pregnancy model as the normal pregnancy group（saline administra原tion）; CBA∕J female mouse and DBA/2 male mouse to produce a spontaneous abortion model, and distributed ran原domly as the spontaneous abortion group（saline）, the cyclosporin group（cyclosporin A）, the low dose group（12g∕kg）, the moderate dose group（24g/kg）, and the high dose group（48g∕kg）, each consisted of 6 mice for a continu原ous administration for 14 days. Then, the pregnant mice were killed, number of embryos in uterus was counted, fe原tal resorption rate were calculated. Expression of Foxp3 and RORγt were detected by Western-blot, TGF-βandTNF-αwere tested by immunohistochemistry. Results （1）Compared to the spontaneous abortion mice, fetal resorp原tion rate of mice from the cyclosporin group, the low, moderate and high dose group decreased significantly（25.0%vs 11.9%, 14.3%, 11.1% and 13.2%. P〈0.05）. There were no difference between all the drug groups and the cy原closporin group, neither between each drug group. （2）Compared to the spontaneous abortion mice, expression ofFoxp3 from the cyclosporin, the moderate and high dose group increased significantly（0.700±0.147 vs 1.223±0.240,1.220±0.241 and 1.383±0.216. P〈0.05）. There were no difference between cyclosporin mice and the moderate andhigh dose group; On the contrary, expression of RORγt decreased significantly（1.456±0.165 vs 0.950±0.2
