摘要
目的检测不同化疗敏感程度的卵巢上皮癌组织中肿瘤干细胞相关表面标志CD105的表达,分析其与化疗耐药及预后的关系。方法 56例卵巢癌手术标本,通过临床随访分为临床铂敏感型和铂耐药型,采用Westernblotting检测卵巢上皮癌组织中CD105蛋白水平的表达情况。结果临床铂耐药的卵巢上皮癌组中,CD105的表达明显高于敏感组(P<0.05);晚期及低分化的卵巢癌组织的CD105表达水平明显高于早期及低高分化的卵巢癌组织(P<0.05)。结论 CD105基因的高表达与卵巢癌化疗耐药及预后相关;下调CD105基因的表达或许是逆转肿瘤侵袭及耐药的新途径。
Objective:The stem cell-related cell surface markers CD105 expression in ovarian epithelial cancer tissues with different chemosensitivity was examined to investigate the association between CD105 expression and chemoresistance. Methods:Tissues were collected from 56 patients who underwent cytoreductive surgery. The tissues were divided into clinical platinum sensitive group and platinum resistant group by clinical follow-up. The expression levels of CD105 in 56 ovarian carcer tissues were determined by Western blotting assay. Results:There were significant differences of CD105 gene expression in epithelial ovarian cancer tissues. The expression of CD105 in clinical platinum resistant ovarian epithelial cancer tissues were significantly higher than that in sensitive tissues(P〈0.01).The advanced-stage(III)or low-differentiated ovarian epithelial cancer showed higher CD105 expression than that of early-stage(I,II)or well-to mild-differentiated ovarian epithelial cancer(P〈0.01). Conclusion:The results indicated that the CD105 overexpression is correlated to chemo-resistance and poor prognosis. Down regulation of CD105 expression may be as a new method in ovarian cancer therapy.
