摘要
目的建立小鼠巨细胞动脉炎(GCA)异种移植模型,以模拟GCA所致的血管炎性损伤过程。方法雄性NOD-scid小鼠32只,随机分为两组:颞动脉移植物+生理盐水组,颞动脉移植物+人外周m单核细胞(PBMC)+生理盐水组,每组16只。将疑似GCA患者颞动脉活检组织(阳性3份,阴性7份)移植到小鼠的腹主动脉前壁,随后尾静脉注射人PBMC。通过对移植后组织切片苏木素-伊红(HE)染色,测量GCA阳性和阴性组织的内膜及中膜厚度并计算其内膜/中膜厚度比率(I/M比率),计数浸润的单核细胞数量。使用免疫组织化学染色定位移植物中的人PBMC细胞。结果在颞动脉移植物+生理盐水组中,GCA阳性移植样本较阴性者I/M比值显著升高(P=0.048);输注了PBMC的GCA阳性移植样本较未输注者发现大量单核细胞浸润(P=0.041)。结论小鼠GCA异种移植模型能较好的反映血管炎性损伤的过程及程度,且该模型操作简便、易于推广。
Objective A xenotransplantation model of giant cell arteritis (GCA) in mice was established to simulate the process of vaseulitis injury induced by GCA. Methods Thirty-two male nod-scid mice were randomly divided into two groups : temporal artery graft + saline group and temporal artery graft + human peripheral blood mononuclear cells (PBMC) + saline group with 16 mice in each group. Temporal artery biopsy tissues (3 positive and 7 negative) of suspected GCA patients were transplanted to the anterior wall of abdominal aorta of mice and then human PBMC was injected into caudal vein. The thickness of intima and media of GCA positive and negative tissues were measured by hematoxylin-eosin (HE) staining after transplantation and their I/M ratio was calculated. Count the number of in- filtrating monocytes, hnmunohistochemical staining was used to locate human PBMC in grafts. Results In the temporal artery graft + saline group, the I/M ratio of GCA positive graft samples was significantly increased than that in the negative ones ( P = 0.048 ) ; a large number of monocyte infiltration was found in GCA positive transplant samples infused with PBMC compared with those without infusion ( P=0.041). Conclusion The xenotransplantation model of GCA in mice can better reflect the process and degree of vasculitis injury, and has the advantages of easy operation and popularization. It is an ideal model to study the progress and treatment of GCA disease.
