摘要
目的观察兔基底尖动脉瘤形成过程中c-Jun氨基末端激酶(JNK)及B细胞淋巴瘤/白血病-2相关X蛋白(bax)的表达。方法新西兰大白兔40只,分为正常组5只,假手术组5只,手术组每组各10只(术后2、8、16周)。采用双侧颈总动脉结扎法造模,假手术组仅暴露双侧颈总动脉,于规定时间行基底动脉尖部磁共振成像(MRA)。所有实验动物处死行病理染色观察基底动脉尖形态、血管内膜及中弹力层变化,免疫组织化学染色。结果免疫组织化学染色结果,JNK在各组表达阳性率分别为对照组:(8.600±2.074)%与术后2周组[(18.900±4.458)%,t=13.405,P=0.000]、术后8周组[(25.800±3.615)%,t=22.570,P=0.000],术后16周组[(38.4004-8.343)%,t=14.555,P=0.000]比较差异有统计学意义。bax的阳性表达率分别为:术后2周组的阳性率为(9.400±3.050)%,与术后2周组[(22.100±4.748)%,t=14.719,P=0.000],术后8周组[(29.200±4.894)%,t=18.866,P=0.000],术后16周组[(36.700±4.762)%,t=24.371,P=0.000]比较差异有统计学意义。影像学结果:对照组动脉瘤体积为(7.600±3.847)mm^3、与术后2周组[(24.746±27.813)mm^3,t=2.814,P=0.020]、术后8周组[(37.083±35.695)mm^3,t=3.285,P=0.009]、术后16周组[(105.751±97.094)mm^3,t=3.444,P=0.007]比较差异有统计学意义。结论JNK与bax参与了兔基底尖动脉瘤的形成过程。
Objective To observe the expression of c-Jun N-terminal kinase(JNK) and B cell lymphoma/leukemia-2 associated X protein (bax) during the formation of basilar aneurysm in the rabbit. Methods Forty New Zealand white rabbits were divided into five group, Normal group (n=5 ), Sham group ( n=5 ) , Operation group ( 2 weeks, 8 weeks, 16 weeks ) ( n=10 ). The bilateral common carotid artery was exposed in the sham operation group by bilateral common carotid artery ligation. MRA (Magnetic Resonance Angiography) was performed at the prescribed time, All experimental animals were killed in order to observe the pathological staining of the basilar artery tip morphology, vascular intima, the middle elastic layer and the immunohistochemieal staining. Results The results of immunohistochemical staining showed that the positive rate of JNK in each group was ( 8. 600±2. 074)% in the control group, (18. 900± 4.458)% in the 2W group (t=13.405, P= 0.000), and the postoperative 8W group (25.800±3. 615)% (t =22. 570, P=0. 000), postoperative 16W group (38. 400±8. 343)% (t = 14. 555, P= 0. 000) ; the difference between the experimental group and the control group was statistically significant.
