肥大细胞表达组织蛋白酶S对类风湿关节炎小鼠CAIA模型关节评分的影响 被引量:3

摘要 目的拟通过C57BL/6小鼠和肥大细胞敲除的Kit^(W-sh/W-sh)小鼠进行胶原抗体诱导关节炎(CAIA)造模,以期发现肥大细胞可能是通过表达组织蛋白酶S对关节症状产生影响。方法选用C57BL/6小鼠和同基因背景的Kit^(W-sh/W-sh)小鼠进行CAIA造模,并对部分肥大细胞敲除小鼠预先尾静脉补充骨髓源性肥大细胞,同时给予肥大细胞激活剂(C48/80)和膜稳定剂(DSCG),计算各组中关节炎评分,用ELISA法检测外周血清中组织蛋白酶S水平,用Western blot方法检测滑膜中组织蛋白酶S的蛋白水平。结果 CAIA模型是一个方便而快捷的类风湿关节炎(RA)模型,可以较好地模拟RA中自身免疫诱发滑膜炎情况。在肥大细胞敲除的Kit^(W-sh/W-sh)小鼠中未成功复制关节炎模型,而提前输注外源性肥大细胞后即出现关节炎。小鼠外周血中组织蛋白酶S的水平与关节炎评分呈显著相关,滑膜部位组织蛋白酶S表达升高。结论肥大细胞表达的组织蛋白酶S是导致关节炎的一个重要因素。 Objective To conduct the CAIA model on C57 BL/6 mice and mast cells knock-out Kit^W-sh/W-sh mice,and thus to find out that mast cells may affect arthritis symptoms by the expression of cathepsin S. Methods CAIA model were conducted on C57 BL/6 mice and Kit^W-sh/W-shmice. The bone marrow-derived mast cells were injected into the Kit^W-sh/W-shmice before experiment. Mast cell' s activator C48/80 and membrane stabilizer DSCG were used in the experiment. The arthritis scores of each group were calculated,and the expression of cathepsin S in peripheral blood was detected. Western blot was used to detect the expression of cathepsin S protein level in the synovial membrane. Results CAIA model was a convenient rheumatoid arthritis( RA) model,which could simulate the immune-induced synovitis in RA. RA model was not conducted in Kit^W-sh/W-shmice,while arthritis was developed by the infusion of exogenous bone marrow derived mast cells. The level of cathepsin S in serum had a good correlation with arthritis scores,so was the expression of Cathepsin S in the synovial membrane. Conclusion Cathepsin S expressed by mast cells might be one of the important factors in causing arthritis.
