枸杞多糖对HepG2肝癌细胞自噬的影响 被引量:3

摘要 目的研究枸杞多糖(LBP)对肝癌细胞自噬的影响。方法体外培养HepG2肝癌细胞,对照组加入不含LBP的等体积培养液,处理组分别加入200、400和800 mg·L-1 LBP,CCK-8法检测各组细胞增殖率。免疫荧光法检测各组微管相关蛋白轻链3(LC3)荧光颗粒数目。Western blot法检测各组LC3II/LC3-I蛋白含量比值变化。结果 LBP对HepG2细胞作用72h后,400、800mg·L-1的LBP与对照组相比,具有抑制细胞增殖的作用(P<0.05);200、400和800 mg·L-1 LBP组作用72h后HepG2细胞中LC3荧光颗粒数目均高于对照组(P均<0.05);400 mg·L-1和800 mg·L-1LBP作用72h后LC3II/LC3-I比值高于对照组(P<0.05)。结论 LBP可抑制细胞的增殖并诱导HepG2细胞自噬。 Objective To investigate the roles of lycium barbarum polysaccharides(LBP) on autophagy of HepG2 cells. Methods HepG2 hepatoma cells were cultured in vitro. The control group was added with LBP-free medium. The treatment group was added with 200,400 and 800 mg·L^-1 LBP respectively. The proliferation rate of each group was detected by CCK-8 method. The number of LC3 fluorescent particles in each group was detected by immunofluorescence. Western blot assay was used to detect the ratio of LC3 II/LC3-I protein content in each group. Results LBP could inhibit the proliferation of HepG2 cells 72 hours after LBP treatment(P〈0.05). After 72 hours of LBP treatment,the numbers of LC3 fluorescent particles in 200,400 and800 mg·L^-1 LBP groups were higher than that in the control group(P〈0.05). The number of LC3 fluorescent particles in 200,400 and 800 mg·L^-1 LBP groups was higher than that in the control group 72 hours after treatment(P〈0.05),and the ratio of LC3 Ⅱ/LC3-I in 400 mg·L^-1 and 800 mg·L^-1 LBP groups 72 hours after treatment was higher than that in the control group(P〈0.05). Conclusion LBP can inhibit the proliferation of HepG2 cells and induce the autophagy to affect the cell survival.
