研究旨在探究miR-181b-5p与Ⅱ型激活素A受体基因(ACVR2A)之间的靶标关系,以及它们对鸡肌肉生长发育的影响。试验过表达或抑制鸡原代成肌细胞中miR-181b-5p及ACVR2A,分析它们与鸡成肌细胞增殖之间的关系,并构建双荧光素酶报告载体确定miR-181b-5p与ACVR2A之间的靶标关系。双荧光素酶活性检测结果显示,共转染miR-181b-5p和pmirGLO-ACVR2A-3′UTR-WT后,miR-181b-5p能显著抑制报告基因活性(P<0.05)。在过表达miR-181b-5p后,ACVR2A的表达量极显著(P<0.01)降低,抑制miR-181b-5p后,ACVR2A的表达量极显著(P<0.01)上升,但无论是过表达还是抑制,对原代成肌细胞的增殖没有影响;在过表达ACVR2A后,miR-181b-5p的表达量极显著(P<0.01)降低,抑制ACVR2A后,miR-181b-5p的表达量极显著(P<0.01)上升。综上所述,miR-181b-5p与ACVR2A之间具有靶标关系,但是miR-181b-5p与ACVR2A对鸡原代成肌细胞的增殖都不具有促进或抑制作用。
The purpose of this study was to explore the relationship between miR-181b-5p and Activin type Ⅱ receptor A gene (ACVR2A)and their relationship with the growth and development of chicken muscles. In this experiment, overexpression and inhibition of miR-181b-5p and ACVR2A were performed on chicken myoblasts, to verify the relationship between them and the proliferation and differentiation of myoblasts, and the target relationship between miR-181b-5p and ACVR2A was verified by a dual-luciferase reporter. After co-transfected with miR-181b-5p and pmirGLO-ACVR2A-3'UTR-WT, miR-181b-5p significantly inhibited the reported gene activity (P〈0.05). After overexpression of miR-181b-5p, the expression of ACVR2A was significantly decreased (P〈0.01), and the expression of ACVR2A was significantly increased after inhibiting miR-181b-5p (P〈0.01), however, neither overexpression nor inhibition had any effect on the proliferation or differentiation of myoblasts. After the overexpression ACVR2A, the expression of miR-181b-5p was significantly increased (P〈0.01), but inhibit ACVR2A, the expression level of miR-181b-5p was significantly decreased (P〈0.01). In conclusion, miR-181b-5p has a target relationship with ACVR2A, but miR-181b-5p or ACVR2A do not promote or inhibit the growth or development of chicken muscles.
