当归补血汤对糖尿病大鼠肝脏、骨骼肌细胞膜上GLUT4的影响 被引量:10

摘要 目的:研究当归补血汤对糖尿病大鼠肝脏、骨骼肌细胞膜上GLUT4表达的影响。方法:选取雄性SD大鼠50只,随机分为空白对照组(NC组)、当归补血汤对照组(DG组)、2型糖尿病模型组(T2DM组)、2型糖尿病+当归补血汤组(T2DM+DG组)、2型糖尿病+吡格列酮组(T2DM+BG组)。采用链脲佐菌素(STZ)结合高脂高糖饮食建立大鼠糖尿病模型(T2DM),造模成功后,药物干预4周后,取血检测大鼠空腹血糖(FPG)、空腹胰岛素(FINS)水平。光镜下观察肝脏病理结构,用免疫组织化学法检测各组大鼠肝脏、骨骼肌细胞膜上GLUT4表达。结果:与NC组比,T2DM组FPG、FINS显著升高。而T2DM+DG组和T2DM+BG组较T2DM组,FPG、FINS下降。光镜下观察到T2DM+DG组和T2DM+BG组肝脏组织得到明显改善。通过免疫组化发现,T2DM组肝脏和骨骼肌细胞膜上GLUT4的表达下降,而T2DM+DG组和T2DM+BG组较T2DM组表达升高,T2DM+DG组与T2DM+BG组组间无明显差异。结论:当归补血汤降糖作用显著,其作用与吡格列酮相当;其降糖作用可能与增强GLUT4蛋白的表达,改善胰岛素抵抗有关。 Objective: Under the treatment of Danggui Buxue Decoction, to observe the GLUT4 expression of diabetic rats' liver and skeletal muscle. Methods: Fifty SD rats were randomly divided into blank group(NC), Danggui Buxue Decoction group(DG), model group(T2 DM), mode+ Danggui Buxue Decoction group(T2 DM +DG),model+pioglitazone group(T2 DM +BG).Using urea with cephalosporins(STZ) combined with high fat and sugar diet,we established rat model of diabetes(T2 DM). After 4 weeks, we measured fasting blood glucose(FPG) and fasting insulin(FINS) in rats. We observed the liver pathology structure and used immunohistochemical method to detect the GLUT4 expression of diabetic rats' liver and skeletal muscle. Results: The FPG and FINS in the T2 DM group were significantly higher than those of the NC group. Compared with the T2 DM group,the data declined in the T2 DM+DG group and the T2 DM+BG group. Under the light microscopy, the liver pathology structure of T2 DM +BG group and T2 DM+BG group got recovered. The T2 DM group had a lower level of the expression of GLUT4 on the liver and skeletal muscle tissue. The T2 DM+DG group and the T2 DM+BG group's were significantly increased. The T2 DM +DG group had no significant difference compared with the T2 DM+BG group. Conclusions: Danggui Buxue Decoction can decrease blood glucose and insulin levels, which is equivalent to pioglitazone. Danggui Buxue Decoction decreases blood glucose,which is related to decreasing the expression of GLUT4 protein and improving insulin resistance.
