摘要
为了验证酶联免疫吸附实验(ELISA)中一种简化的的氯霉素包被方法,采用间接竞争酶联免疫吸附法(icELISA)对氯霉素标准品和牛乳样品进行了测定,分析了测量孔间吸光度值变异系数,制备了吸光度值与氯霉素浓度关系的回归曲线,根据回归曲线测算出牛乳样品中氯霉素检出量和回收率,并与目前通用的氯霉素包被方法进行了对比。研究发现,在其它条件一定的情况下,这种简化的包被方法与通用的包被方法相比回归曲线具有更大的斜率,吸光度值的变异系数更低,两者在牛乳样品中氯霉素检出量和回收率方面没有显著性差异。结果表明,这种包被方法可以获得理想的包被效果,能够用于样品中氯霉素的检测。
In order to verify a simplified method of chloramphenicol coating in enzyme linked immunosorbent assay (ELISA), indirect com-petitive enzyme-linked immunosorbent assay (icELISA) was used to determine chloramphenicol standards and milk samples, the coefficient of variation of ahsorhance between holes was measured, the regression curve of the relationship between the ahsorhance and the concentra- tion of chloramphenicol was prepared, the detection amount and recovery rate of chloramphenicol in milk samples were calculated according to the regression curve, and we compare these results with the current common coating method of chloramphenicol. It is found that, in the case of the same other conditions, the simplified coating method has a greater slope in the regression curve and lower variation coefficient of the ahsorhance compared with the common coating method, however, there was no significant difference in the detection amount and recov- ery rate of chloramphenicol in the milk samples between the two methods. The results show that the coating method has a satisfactory effect and can be used for the detection of chloramphenicol in the sample.
