摘要
目的分析并归纳在脊髓缺血再灌注(SCI/R)过程中TLR-4的表达与血清及脑脊液中炎性细胞因子的水平。方法建立脊髓缺血再灌注动物模型,在缺血再灌注的不同时间点来检测血清中TNF-α、IL-6及脊髓组织中TLR-4的表达分布情况。结果实验组TNF-α、IL-6表达水平均较对照组升高(P均<0. 05),随着缺血时间的增加,TNF-α、IL-6表达水平是逐渐上升的;与对照组比较,随着缺血时间的延长,TLR-4水平逐渐增加(P <0. 01)。结论在大鼠缺血再灌注模型中,TNF-α、IL-6、TLR-4表达量增加,它们参与SCI/R病理过程。
Objective To study the expression of TLR-4 and the levels of inflammatory eytokines in serum and eerebrospinal fluid after spinal isehemia-reperfusion in rats. Methods An animal model of spinal isehemia-reperfusion was established. The levels of TNF-α andIL-6 in serum were quantified by ELISA kits at different time points after ischemia-reperfusion in rats, and the expression of TLR-4 mRNA and protein was detected with RT-PCR and immunohistoehemical staining, respectively. Results The levels of TNF-α, IL-6 in serum and the expression of TLR-4 mRNA and protein in spinal tissues were gradually increased with the increase of ischemie time,and there had significant difference as compared with the control group (P 〈 0.01 ). Conclusion The high expression of TNF-α ,IL-6 and TLR-4 were involved in the spinal cord isehemia-reperfusion injury.
