基于超高效液相色谱-四极杆/静电场轨道阱高分辨质谱技术的脑心通胶囊化学成分研究 被引量:11

The Chemical Constituents Study of Naoxintong Capsules Based on UHPLC-Q-Orbitrap HRMS

摘要 目的为系统研究中药复方制剂脑心通胶囊的化学成分组成,本实验采用先进的超高效液相色谱-四极杆/静电场轨道阱高分辨质谱(UHPLC-Q-Orbitrap HRMS)技术建立一种快速准确识别脑心通胶囊中多种化学成分的定性分析方法。方法本实验通过分析UHPLC-Q-Orbitrap HRMS技术捕捉未知化合物的一级精确相对分子质量和多级碎片信息,同时与对照品的保留时间和质谱数据信息进行比对,并结合相关参考文献以及Mass Bank、Chemical Book等网络数据库信息从而实现对药物中化学成分的准确快速定性。结果本实验从脑心通胶囊中共鉴定出44种化学成分,其中主要包括黄酮类、醌类、萜类、及有机酸类等化学成分。结论本实验建立的方法可快速、准确,系统地识别脑心通胶囊中多种化学成分,并为其质量控制、药效物质基础研究及进一步的临床应用提供坚实的理论依据和科学的研究思路。 OBJECTIVE To establish a novel ultra-high performance liquid chromatography-quadrupole/orbitrap high resolution mass spectrometry (UHPLC-Q-Orbitrap HRMS) method to study the chemical composition of Naoxintong capsules rapidly and accu- rately. METHODS The information of accurate mass in full-MS and multistage fragment ions was obtained by the novel UI-IPLC-Q- Orbitrap technology. Chemical constituents were identified by comparing the relative retention time and the mass data of the reference substances, meanwhile consulting the reference literature or the Mass Bank, Chemical Book network database. RESULTS Forty- four compounds were finally identified in this study, mainly including flavones, authraquinones, terpenoids and organic acids. CONCLUSION A method is established in this study to identify various chemical constituents of Naoxintong capsules rapidly, ac- curately and systematically. What's more, our research will lay a sound theoretical foundation and propose a scientific study idea for the quality control improvement, bioactive components recognition and further clinical application of this herbal formulation.
