摘要
循环游离DNA(cell-free DNA,CF-DNA)可由正常细胞和肿瘤细胞释放,其中肿瘤细胞来源的游离DNA又叫做循环肿瘤DNA(circulating-tumor DNA,CT-DNA),以细胞外游离形式存在于血液、尿液等体液中。在前列腺癌(PCa)患者中,CF-DNA的浓度明显高于健康对照组,而且存在与原位肿瘤相似或完全相同的基因突变、超甲基化、基因不稳定性等变化特征,对肿瘤的早期诊断、术后检测以及对靶向药物的疗效评估都有着潜在作用,是新兴的肿瘤标志物研究热点。本综述简要阐述CF-DNA的生物学特性、检测以及作为肿瘤标志物的新进展。
Cell-free DNA(CF-DNA)can be released from normal cells and tumor cells.The free DNA from the tumor cell is called circulating tumor-DNA(CT-DNA).It exists in the blood,urine or other body fluids in the form of extracellular dissociation.In prostate cancer,the concentration of CF-DNA is significantly higher than that in the healthy control group,and there are changes in gene mutation,hypermethylation and gene instability,similar or identical to the tumor in situ.It has a potential role in the early diagnosis of the tumor,the post operation test,and the evaluation of the efficacy of the targeted drug,a new research hotspot of tumor markers.This review briefly describes the biological characteristics of circulating CF-DNA,and its detection and new progress as a tumor marker.
