摘要
目的研究丹皮酚与丹参酮ⅡA联合重组人粒细胞刺激因子(rhG-CSF)对大鼠脑缺血损伤的保护作用,并初步探讨其机制。方法采用制备大鼠局灶性脑缺血(MCAO)模型,用丹皮酚与丹参酮ⅡA联合rhG-CSF进行干预,检测用药后的神经功能评分、脑梗死率和含水率;采用Western Blot法检测大鼠海马血管生成素1(Ang(-1))、酪氨酸激酶受体2(Tie-2)和整合素(integrin)α6、β1、α5蛋白的表达。结果与模型组比较,丹皮酚与丹参酮ⅡA联合rhG-CSF明显改善了大鼠的神经功能缺损症状评分、降低脑梗死率和含水率;上调了大鼠海马Ang(-1)、Tie-2、integrinα6、integrinα5蛋白的表达。结论丹皮酚与丹参酮ⅡA联合rhG-CSF对脑缺血大鼠的保护作用可能与Ang(-1)/Tie-2、integrinα6等调控神经血管再生的关键分子蛋白的激活有关。
OBJECTIVE To study the protective effects of paeonol and salviolⅡA combined with rhG-CSF on the focal cerebral ischemia in rats and the mechanism. METHODS The cerebral ischemia model was induced by MCAO. To investigate the neuroprotective effect of paeonol and salviolⅡA combined with rhG-CSF,the neurological deficit scores,cerebral infarct size and water content were determined after MCAO. The protein expression levels of angiopoietin(-1)( Ang(-1)),tyrosine kinase receptors 2( Tie-2),integrin α6,integrin β1 and integrin α5 were analyzed by Western Blot method. RESULTS Paeonol and salviolⅡA combined with rhG-CSF significantly ameliorated the neurological deficit,greatly reduced the infarct size and water content. Compared with the model group,the protein expression level of Ang(-1),Tie-2,integrin α6 and integrin α5 in hippocampus were upregulated by paeonol and salviolⅡ A combined with rhG-CSF. CONCLUSION The protective effect and the mechanism of paeonol and salviol Ⅱ A combined with rhG-CSF treatment may be related to the activation of Ang(-1) and Tie-2 signaling pathway and the integrin α6. These factors play an important part in neuro protection and angiogenesis after cerebral ischemia.
