摘要
生物硫醇的定量和动态变化的检测一直是难点问题。基于点击化学的高效、可靠、高选择性的特点和化学试剂的易得性,选择醇羟基和酚羟基化合物进行了探讨研究。以醇羟基苯甲醇为原料,用丙烯酰氯酰化,与L-半胱氨酸进行巯基-双键反应,得到标题化合物,其结构经核磁共振谱、质谱和红外光谱确定,总收率89%,可以作为半胱氨酸荧光探针设计的依据。而以酚羟基、苯酚和8-羟基喹啉为原料,得到七元环的分子内氨解产物,是半胱氨酸紫外检测的设计基础。
The quantitative and dynamic detection of biothiols has always been a difficult problem.Based on the characteristics of high efficiency,reliability and selectivity of click chemistry and the availability of chemical reagents,alcohol hydroxyl and phenolic hydroxyl compounds being selected was studied.The title compound was obtained by thiol-ene reaction of L-cysteine with alcohol hydroxy benzyl alcohol and acroleyl chloroacylation,and the structure was determined by NMR,MS and IR. The total yield is89%,which can be used as a basis for designing cysteine fluorescent probes.Meanwhile,the intramolecular ammonolysis product of the seven membered ring was obtained by using phenolic hydroxyl,phenol and 8-hydroxyquinoline as raw materials,which is the design basis of cysteine ultraviolet probes.
