摘要
目的:探讨恒古骨伤愈合剂(osteoking,OK)通过激活Wnt/β-catenin信号通路增加骨形成缓解OP的作用机制。方法:50只雌性树鼩随机分为5组,正常对照组正常喂养,OP模型组去除双侧卵巢每天进行生理盐水灌胃,OK低、中、高剂量组分别给予OK 0.5、1.5、3.0 mL/d灌胃。10周后,检测骨密度(BMD);处死树鼩,取血清和骨骼,ELISA法测定血清雌二醇(E_2)、骨碱性磷酸酶(BALP)、骨钙素(OC)、Ⅰ型原胶原N-端前肽(PINP)、抗酒石酸酸性磷酸酶5b(TRACP-5b)和Ⅰ型胶原交联C-末端肽(CTX)的含量;采用RT-q PCR和Western blot法检测骨组织Wnt1和β-catenin的表达水平。结果:与正常对照组比较,模型组树鼩血清E2含量和BMD下降明显(P<0.05),CTX、TRACP-5b、OC、PINP和BALP的含量均显著上升(P<0.05),经OK治疗后上述现象均改善明显,且具有剂量依赖性;RT-q PCR与Western Blotting结果显示Wnt1和β-catenin的表达水平经OK治疗后均明显升高。结论:OK能够通过增加E_2分泌、抑制骨吸收和促进骨形成增加骨密度缓解OP,其作用机制可能是通过激活Wnt/β-catenin信号通路实现的。
Objective: To explore the metabolism of enhancing bone formation and ameliorating OP of OK via the Wnt/β-catenin signal pathway in the castration OP tree shrew models. Methods: 50 female experimental tree shrews were randomly divided into normal control group,OP model group animals were removed bilateral ovaries and received saline by gavage daily,the tree shrews in OK-L,OK-M and OK-H groups were given OK 0. 5,1. 5 and 3. 0 mL/d by gavage for 10 weeks,determining BMD,all the tree shrews were sacrificed and serum and bone were collected,ELISA was employed to determine the concentrations of E_2,BALP,OC,PINP,TRACP-5 b and CTX; RT-q PCR and Western blotting were used to detect the expression levels of Wnt1 and β-catenin. Results: Compared with the normal control group,the serum E_2 level and BMD decreased significantly( P〈0. 05) in the OP group,CTX,TRACP 5 b,OC,PINP and BALP levels were significantly increased( P〈0. 05),after intervened with OK,all the above indexes were improved obviously,and the effect took the shape of a dose-dependent manner; RT-q PCR and Western Blotting results showed that the expressions of Wnt1 and β-catenin were up-regulated after intervened with OK. Conclusion: OK can increase the BMD to alleviate OP by raising the secretion of E_2,inhibiting bone resorption and promoting bone formation, the mechanism is relate to the activation of Wnt/β-catenin signaling pathway probably.
