摘要
为了研究黄芩提取物对单增李斯特菌溶血素(LLO)溶血活性及单增李斯特菌致病力的影响,试验采用测定最小抑菌浓度(MIC)、绘制生长曲线、分析溶血活性、蛋白免疫印迹分析和细胞毒性试验的方法分析了黄芩提取物对单增李斯特菌增殖、单增李斯特菌溶血素溶血活性和单增李斯特菌对细胞的毒性作用。结果表明:黄芩提取物可通过直接中和单增李斯特菌溶血素活性而降低溶血活性,且这一作用与黄芩提取物的抗菌活性无关;在细菌与细胞共培养体系内加入黄芩提取物可有效抑制细菌对细胞介导的细胞毒性作用。说明黄芩提取物是一种潜在的抗单增李斯特菌感染的先导化合物。
In order to determine the effect Scutellaria baicalensis Georgi exact on the activity of listeriolysin O and the pathogenicity of L. monocytogenes,bacterial proliferation,pore forming activity of listeriolysin O and cytotoxicity mediated by L. monocytogenes in the presence or absence of Scutellaria baicalensis Georgi exact were analysized. Here,it used MIC assay,growth assay,hemolysis assay,Western-blot and cytotoxicity test to investigate the effect of Scutellaria baicalensis Georgi exact on the pore-forming ability of L. monocytogenes. The results showed that the Scutellaria baicalensis Georgi exact could inhibit the hemolytic activity of bacterial culture supernatants following co-cultured with L.monocytogenes by directly neutralizing LLO function. Furthermore,the addition of Scutellaria baicalensis Georgi exact into the co-culture system of L. monocytogenes and J774 could significantly attenuate the cell injury induced by L. monocytogenes. Our results established that Scutellaria baicalensis Georgi exact may represent a promising leading compound for L. monocytogenes infection.
