摘要
6-磷酸果糖-2-激酶/果糖-2,6-二磷酸酶4(PFKFB4)是癌细胞糖代谢途径中的关键酶,也是双功能酶6-磷酸果糖-2-激酶/果糖-2,6-二磷酸酶(PFKFB)的四个亚型之一,主要功能是合成磷酸果糖激酶-1(PFK-1)的最强变构激活剂果糖-2,6-二磷酸(F-2,6-BP),控制糖酵解通量的进行及促进ATP的生成,从而影响肿瘤生长,PFKFB4是多种肿瘤细胞生存所必需的。近年来PFKFB4逐渐成为靶向药物研究的新热点,该文从PFKFB4在肿瘤糖代谢中的作用及其在肿瘤形成、进展及预后等方面的分子机制作出综述。
6-phosphofructo-2-kinase/fructose-2, 6-biphosphatase 4(PFKFB4) is a key enzyme in the pathway of glucose metabolism, which is one of the four subtypes of the bifunctional enzyme 6-phosphofructo-2-kinase/fructose-2, 6-biphosphatase(PFKFB). The main function of PFKFB4 is to synthesize the strongest allosteric activator of phosphoric acid fructose kinase -1(PFK-1), fructose-2, 6-bisphosphate(F-2, 6-BP). PFKFB4 regulates glycolysis flux and promotes the production of ATP, which affects the growth of tumors. Studies have shown that PFKFB4 is essential for the survival of many kinds of tumor cells. In recent years, PFKFB4 has gradually become a new focus on targeted medical therapy. In this paper, we review the roles of PFKFB4 in tumor glucose metabolism and its molecular mechanisms in tumorigenesis, progression and prognosis.
