冠状动脉粥样硬化代谢组学生物标志物的筛选 被引量:4

Identification of biomarkers for atherosclerosis by metabolomics

摘要 目的利用代谢组学方法寻找对诊断冠状动脉粥样硬化有潜在临床价值的代谢组学生物标志物。方法收集43例冠状动脉粥样硬化患者和49例健康体检者的血浆样品,利用快速高分辨率液相色谱/高分辨飞行时间质谱联用仪(RRLC-QTOF/MS)对血浆样品进行检测,获得冠状动脉粥样硬化组及健康对照组血浆的代谢指纹图谱,采用统计学中主成分分析法及偏最小二乘判别分析方法来分析冠状动脉粥样硬化组及健康对照组之间的代谢产物的差异,进而筛选出具有潜在诊断意义的代谢组学生物标志物。结果冠状动脉粥样硬化与正常对照组之间的血浆代谢组学对比研究表明,冠状动脉粥样硬化患者与正常对照组的患者能够进行准确地区分,两组在血浆代谢模式上存在着较明显的差异,本研究共筛选鉴定出18个标志物。结论利用代谢组学方法能够对冠状动脉粥样硬化和正常对照组患者较准确区分,鉴定出的标志物对动脉粥样硬化的诊断有潜在价值。 Objective To provide more useful proof on the diagnosis of atherosclerosis by using rapid resolution liquid chromatography quadrupole time-of-flight mass spectrometry(RRLC-QTOF/MS). Methods Forty-three patients with coronary atherosclerosis and 49 cases of healthy check-up of plasma samples were collected,plasma samples were tested by using fast high-resolution liquid chromatography/high resolution time of flight mass spectrometry instrument(RRLC-QTOF/MS). Plasma metabolic fingerprint of coronary atherosclerosis group and healthy control group were obtained,the differences of metabolites between coronary atherosclerosis group and healthy control group were analyzed by the principal component analysis and partial least squares discriminant analysis method,and then biomarkers of metabonomics with potential diagnostic significance were selected. Results There were obvious metabolic differences between atherosclerosis group and normal control group by plasma metabolomic study. Eighteen biomarkers were found. Conclusion The methods of plasma metabolomics based on RRLCQTOF/MS could distinguish clearly. Identified biomarkers have great potential in diagnosis of atherosclerosis patients from normal ones.
