大黄对胆汁淤积幼龄大鼠的预防保护作用 被引量:4

Preventive and Protective Effect of Rhubarb in Young Rats with Intrahepatic Cholestasis

摘要 目的:探讨大黄对胆汁淤积幼龄大鼠的保护效果。方法:将42只幼龄大鼠随机分为正常组、模型组、熊去氧胆酸(UDCA)组,地塞米松(DEX)组,大黄低、中、高剂量组,每组6只,正常组和模型组给予羧甲基纤维素钠溶液灌胃,其余组分别给予UDCA、DEX、及大黄灌胃。第5天各组动物(正常组除外),采用α-萘异硫氰酸酯(ANIT)50 mg/(kg·d)一次性灌胃建立肝内胆汁淤积模型,ANIT灌胃后48 h分别处死各组动物。比色法检测总胆红素(TBIL)、直接胆红素(DBIL)、总胆汁酸(TBA)、碱性磷酸酶(ALP)、γ谷氨酰转移酶(GGT)、丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)。采用苏木精-伊红染色于光镜下观察各组肝脏形态学改变。结果:模型组血清TBIL、DBIL、TBA、ALP、GGT、ALT、AST水平较正常组明显升高(P〈0.01)。UDCA组血清TBIL、TBA、ALP、GGT、AST水平较模型组有不同程度降低(P〈0.01,P〈0.05)。DEX组血清TBIL、TBA、AST水平较模型组有不同程度降低(P〈0.01,P〈0.05)。大黄各剂量组血清TBIL、DBIL、TBA、ALP、GGT、ALT、AST水平较模型组有不同程度降低(P〈0.01,P〈0.05)。大黄高剂量组血清TBIL、DBIL、TBA、ALP、GGT、ALT、AST水平较UDCA组有不同程度降低(P〈0.01,P〈0.05)。大黄中剂量组血清TBIL、TBA、ALP、GGT、ALT、AST水平较UDCA组明显降低(P〈0.01)。大黄低剂量组血清TBIL、TBA、ALT、AST与UDCA组有不同程度降低(P〈0.01,P〈0.05)。大黄不同剂量组血清TBIL、DBIL、TBA、ALP、GGT、ALT、AST水平较DEX组有不同程度降低(P〈0.01,P〈0.05)。大黄低剂量组血清DBIL,TBA,ALP,ALT,AST水平较中剂量组、高剂量组有不同程度升高(P〈0.05,P〈0.01)。中剂量组血清DBIL,TBA,ALP,ALT,AST水平较高剂量组无显著性差异(P〉0.05)。模型组大鼠肝脏病理改变最重,UDCA组、DEX组、大黄不同剂量组肝脏病理 Objective: To explore the protective effect of rhubarb in intrahepatic cholestasis of young rats. Methods: A total of 42 young Sprague-Dawley rats were divided into control group,model group,ursodeoxycholic acid(UDCA) group,dexamethasone(DEX) group,low-dose rhubarb group,medium-dose rhubarb group and high-dose rhubarb group. The rats in control group and model group were given gavage with sodium carboxymethyl cellulose,while the intervention group was given UDCA,DEX and rhubarb solution by gavage. The rats' model of intrahepatic cholestasis was established using [ANIT,50 mg/(kg·d) ] gavage.And the rats in each group were executed 48 h after ANIT gavage. The total bilirubin(TBIL),direct bilirubin(DBIL),total bile acid(TBA),alkaline phosphatase(ALP),gamma-glutamyltranspeptidase(GGT),alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were detected using colorimetry method. Hematoxylin-eosin staining was applied to observe the morphological changes of the liver under a light microscope. Results: Compared with control group,the levels of liver function including TBIL,DBIL,TBA,ALP,GGT,ALT and AST were higher significantly in the model group(P〈0. 01). The levels of TBIL,TBA,ALP,GGT and AST in UDCA group,TBIL,TBA and AST in DEX group and TBIL,DBIL,TBA,ALP,GGT,ALT and AST in different doses of rhubarb were decreased in different degrees(P〈0. 01,P〈0. 05) in contrast with model group. The levels of TBIL,DBIL,TBA,ALP,GGT,ALT and AST in high-dose rhubarb group. TBIL,TBA,ALP,GGT,ALT and AST in medium-dose rhubarb group and TBIL,TBA,ALT and AST in low-dose rhubarb group were decreased in different degrees(P〈0. 01,P〈0. 05) in contrast with UDCA group. The levels of TBIL,DBIL,TBA,ALP,GGT,ALT and AST in different doses of rhubarb were decreased in different degrees(P〈0. 01,P〈0. 05) than those of the DEX group.
