摘要
目的:从肝细胞线粒体角度,探讨归脾汤对雷公藤醇提物致大鼠急性肝损伤的保护机制。方法:30只大鼠随机分为正常组、模型组、归脾汤组3组,每组10只。归脾汤组灌胃9 g/kg归脾汤,连续4 d,再以3.25 mg/kg雷公藤醇提物灌胃模型组、归脾汤组3 d,构建肝损伤模型。测定各组肝组织线粒体中还原型谷胱甘肽过氧化物酶(GSH)、丙二醛(MDA)、超氧化物歧化酶(SOD)水平、三磷酸腺苷酶(ATP酶)含量以及肝细胞线粒体膜电位(△Ψm)水平。结果:与正常组比较,模型组大鼠SOD、GSH、Na^+-K^+-ATPase、Ca^(2+)-Mg^(2+)-ATPase、肝细胞△Ψm水平明显降低,MDA水平则明显升高(P<0.01);与模型组比较,归脾汤组SOD、GSH、Na^+-K^+-ATPase、Ca^(2+)-Mg^(2+)-ATPase、肝细胞△Ψm水平明显升高(P<0.01),MDA水平则明显降低(P<0.01)。结论:归脾汤对雷公藤醇提物致肝损伤大鼠有一定保护作用,其机制与减轻肝细胞线粒体损伤有关。
Objective：To study the protective effect of Guipi decoction on hepatocyte mitochondria in liver injury rat induced by extract from Tripterygiumwilfordii by Ethanol. Method：30 rats were randomly divided into control group,model group,Guipi decoction group. The rats in Guipi decoction group were perfused with Guipi decoction 9 g/kg. After 4 days,control group and the model group was perfused with T. wilfordii 3.25 mg/kg for 3 days. The contents of SOD、GSH、MDA and activity of ATPase of hepatocyte mitochondria were measured. Meanwhile,the mitochondrial membrane potential was observed. Results：Compared with control group,the activities of SOD、GSH、Na＋-K＋-ATPase、Ca^（2＋）-Mg^（2＋）-ATPase、△Ψm in model group were reduced,the content of MDA of mitochondria was increased（P〈0.01）. Compared with model group,activities of SOD、GSH、Na＋-K＋-ATPase、Ca^（2＋）-Mg^（2＋）-ATPase、△Ψm in Guipi decoction group were enhanced,content of MDA of mitochondria was reduced（P〈0.01）. Conclusion：Guipi decoction has protective effect on the liver induced by extract from Tripterygiumwilfordii by ethanol and its mechanism may be related to alleviating mitochondria injury.
