摘要
目的探讨两种Schiff试剂配制方法以及应用于实验教学中的染色效果对比。方法分别采用传统配制方法和改良冷配法配制Schiff试剂。热配法:将碱性品红溶于沸水,并趁热过滤,向滤液中加入盐酸以及亚硫酸氢钠,4℃避光静置过夜。次日加入活性炭,振荡脱色,过滤后即得。冷配法:将碱性品红直接加入一定浓度的盐酸中后加入亚硫酸钠,充分振荡溶解即得。采用蟾蜍全血涂片的Feulgen反应,比较两种方法配制的染液的染色效果。结果两种方法配制的Schiff试剂均将蟾蜍红细胞的细胞核染成紫红色,染色效果相当。结论Schiff试剂改良冷配法操作简便快捷,染色结果清晰,更适于实验教学活动中。
Objective To investigate two kinds of formula of Schiff reagent and compare the application effect in experimental teaching. Methods Schiff reagents were formulated by traditional formula and modifed cold formula. Traditional formula: The basic fuchsin was dissolved in boiling water and filtered. Hydrochloric acid and sodium bisulfite were added into the fltrate. Kept in 4℃ for a night. The next day the activated carbon was added. Finally got the Schiff reagent after filtered. Modified cold formula: the basic fuchsin and sodium sulfte were dissolved in a certain concentration of hydrochloric acid in proper order, and then got the Schiff reagent after oscillated and dissolved. The Feulgen reaction in toad blood smears was used to compare the two formulas. Results The nucleus of toad red cells were stained to fuchsia by two formulas of Schiff reagent with the same depth. Conclusion The improved formula of Schiff reagent is easy and fast to operate and clear in staining, which is more suitable for experimental teaching.
