摘要 采用高效液相色谱柱前衍生法测定酵母抽提物中谷氨酸的含量,以苯异硫氰酸作为柱前衍生试剂,在C18柱上,弱碱性条件以保留时间定性、外标法定量。结果表明,在0.2~1.0mg/mL线性范围内,线性相关系数为0.999816,相对标准偏差小于5%,回收率在95%~110%,样品检测结果良好。该方法还可以用于其他氨基酸的检测。 The content of glutamic acid in yeast extract was determined by high pefformance liquid chromatography column derivatization. Benzo thiocyanic acid was used as a pre column derivatization reagent. On the C 18 column, the retention time was qualitative and the external standard was quantified under the condition of weak alkali. The resuits show- that in the linear range of 0.2 - 1.0mg/mL, the linear correlation coefficient is 0.999816, the relative standard deviation is less than 5%, the recovery rate is between 95% - 110%, and the sample test results ale good. This method can also be used for the detection of other amino acids.
