摘要
利用SRAP标记技术研究45份结缕草属植物的遗传变异情况。从100对引物中筛选出13对扩增带型清晰、多态性较好的引物对其进行扩增,共扩增出113条条带,平均每对引物扩增出8.69条条带,多态性条带数为96条,多态性条带所占比例为84.16%,遗传相似性范围为0.54~0.96,平均为0.75。基于UPMAG(unweighted pair-group method with arithmetic means)法,在遗传相似性系数为0.74时,可将45份结缕草属种质分为4个类群,生态地理条件相似的种质优先聚在一起。本研究结果说明结缕草属植物种质间遗传变异比较广泛,为结缕草属植物种质资源的开发利用及新品种选育奠定了良好基础。
Sequence-related amplifed polymorphism(SRAP) was used to detect the genetic variation and genetic ralationship among 45 Zoysia willd. accessions. A total of 13 primer pairs from the original 100 primer pairs were selected based on the polymorphic, clear and reproducible bands produced by them among 3 Zoysia accessions. A total of 113 bands were amplified by 13 SRAP primer pairs from 45 Zoysia accessions, and 96(84.16%) out of them were polymorphic. The genetic similarity coeffcients(GSC) among the 45 accessions ranged from 0.54 to 0.96 with an average of 0.75. Cluster analysis conducted by the unweighted pair-group method with arithmetic averages(UPGMA) separated the 45 Zoysia accessions into 4 distinct groups when the genetics similarity coefficient was 0.74. Results obtained clearly indicated significant variation existed in the wild Zoysia genotypes, which would provide a theoretical foundation for development and utilization of Zoysia germplasm resources and new variety breeding.
