摘要
目的构建表达干扰迷走神经活性基因的载体靶向转导至犬心房,监测心房肌易损性指标的变化,验证其疗效。方法 17只实验动物随机分为3组:假手术组(n=5):经胸心房肌注射500μL生理盐水;rAd-EGFP+房颤组(n=6):经胸心房肌内注射500μL携带EGFP(绿色荧光蛋白)基因的重组腺病毒溶液。rAd-Gαi2ctp+房颤组(n=6):经胸心房肌内注射500μL携带Gαi2ctp基因的重组腺病毒溶液。7d后检测3组犬左、右心房离体微电极阵列芯片(microelectrode arrays chip,MEA)电传导图和场电位时程(field action potential duration,FAPD)变化,实验结束后收取心房肌行PAS染色检测。结果 rAd-EGFP+房颤组左、右心房组织MEA检测场点位(FAPD)与假手术组和rAd-Gαi2ctp+房颤组比较,差异均有统计学意义(P<0.05),但假手术组和rAdGαi2ctp+房颤组比较,左、右心房组织FAPD差异无统计学意义。3组犬左右心房离体MEA电传导图显示rAdEGFP+房颤组传导紊乱,余2组传导未见明显异常。假手术组与rAd-Gαi2ctp+房颤组左、右心房肌PAS特染组间比较差异无统计学意义(P>0.05)。rAd-EGFP+房颤组与rAd-Gαi2ctp+房颤组和假手术组比较,左、右心房肌PAS特染差异有统计学意义(P<0.05)。结论重组腺病毒转导Gαi2ctp可选择性去迷走、改变心房肌易损性特性,逆转重构的发生发展。
Objective To evaluate the efficacy of Gαi2 ctp gene transduction on the atrial muscle vulnerability,we construct a vector that could express Gαi2 ctp gene,and targeted transduction to the left atrium.Methods Adult beagle dogs were randomly assigned to 3 groups:a sham operation group(n =5),a fast left atrial appendage 6-hr pacing+rAd-EGFP injection group(n =6),and a 6-hr pacing+rAd-Gαi2 ctp injection group(n =6).All samples of left and right atrial tissue were removed and microscopically examined.7 days later,field action potential duration(FAPD)of left and right atrial tissue from each group were measured by microelectrode arrays chip(MEA).Atrial muscle was stained by PAS.Results There was significant difference in FAPD between the rAd-EGFP+AF group and the sham-operated group and the rAd-G alpha i2 ctp+AF group(P 0.05),but there was no significant difference in FAPD between the sham-operated group and the rAd-G alpha i2 ctp+AF group.MEA electrical conduction map of left and right atria in three groups showed conduction disorder in rAd-EGFP+AF group,and there was no significant abnormality in the other groups.There was no significant difference between sham operation group and rAd-G alpha i2 ctp+atrial fibrillation group(P 0.05).Compared with rAd-G alpha i2 ctp + AF group and sham operation group,PAS staining of left and right atrial muscle had significant difference(P0.05).Conclusion rAd-Gαi2 ctp could selectively interferes with the vagus nerve effect and change the vulnerability of atrial muscle,reverse atrial remodeling in dogs.
