细胞凋亡抑制基因和促凋亡基因在喉鳞癌组织中的表达与临床意义 被引量:2

Expression and its significance of apoptosis suppressor gene and proapoptotic gene in laryngeal squamous cell carcinoma

摘要 目的研究细胞凋亡抑制基因(bcl-2)、促凋亡基因(Bax)在喉鳞癌中的表达情况及其临床意义。方法收集2017年1月至7月在本院确诊的54例喉鳞癌患者的癌组织和对应的癌旁组织,用免疫印迹法测定这2种组织中Bcl-2蛋白、Bax蛋白的表达情况;用免疫组化(SP)法测定Bcl-2蛋白、Bax蛋白与血管内皮生长因子-C(VEGF-C)蛋白水平及其阳性表达情况,探讨Bcl-2蛋白、Bax蛋白表达与喉鳞癌患者临床病理特征之间的关系,分析Bcl-2蛋白、Bax蛋白的表达与VEGF-C表达的关系,探讨Bcl-2蛋白、Bax蛋白阳性表达病例在这2种组织中微血管密度(MVD)间的差异。结果免疫印迹法测得:喉鳞癌患者癌组织中的Bcl-2蛋白、Bax蛋白表达明显高于对应癌旁正常组织(P<0.05)。SP测得:Bcl-2蛋白、Bax蛋白、VEGF-C蛋白在喉鳞癌患者癌组织中的表达阳性率分别为53.7%,50%,42.6%,明显高于癌旁正常组织,差异均有统计学意义(均P<0.05)。喉鳞癌患者癌组织中Bcl-2蛋白、Bax蛋白表达阳性率跟喉鳞癌临床分期、喉鳞癌复发或转移、淋巴结转移均相关(均P<0.05)。经Pearson相关性分析,喉鳞癌患者癌组织中Bcl-2蛋白、Bax蛋白的表达均与VEGF-C表达呈正相关(r=0.762,0.703;均P<0.05)。在癌组织中,Bcl-2蛋白、Bax蛋白阳性表达病例MVD值分别为56.21±1.76,52.05±1.31;均明显高于癌旁正常组织中MVD值(30.83±1.45,29.93±1.27),差异均有统计学意义(均P<0.05)。在癌组织中,Bcl-2蛋白、Bax蛋白阴性表达病例MVD值分别为31.03±2.14,30.16±1.95,与癌旁正常组织中的MVD值比较差异无统计学意义(均P>0.05)。结论喉鳞癌组织中Bcl-2蛋白和Bax蛋白存在异常表达,其可能会促进VEGF-C表达,以诱发微血管在喉鳞癌癌组织中的新生。 Objective To investigate the relationship between apoptosis related genes( bcl-2),proapoptotic gene( Bax) and the occurrence and development of laryngeal squamous cell carcinoma( LSCC),and to investigate the correlation between Bcl-2 and Bax protein and angiogenesis. Methods From January 2017 to July,54 cases of LSCC tissue and paracancerous tissues were colected. The expression of Bcl-2 and Bax protein in LSCC samples were determined by Western blotting in cancer tissue. The expression and its positive expression of Bcl-2,Bax,vascular endothelial growth factor-C protein( VEGF-C) levels were determined by immunohistochemical. The relationship between Bcl-2,Baxexpression and the clinicopathological characteristics of patients with LSCC was investegated. The relationship between expression of VEGF-C and Bcl-2,Bax protein expression was analyzed. The difference of the microvessel density( MVD) of cases of laryngeal squamous cancer and the expression of Bcl-2 and Bax protein in the two tissues were investegated. Results The expression of Bcl-2 and Bax protein in cancer tissues of LSCC patients was significantly higher than that in adjacent normal tissues with Western blotting( all P〈 0. 05). The positive rates with SP of Bcl-2,Bax and VEGF-C protein in LSCC patients were 53. 7%,50% and 42. 6%,which were significantly higher than those in normal tissues( all P〈 0. 05). The positive rates of Bcl-2 and Bax protein in LSCC patients were correlated with clinical stage,recurrence or metastasis and lymph node metastasis with significantly( all P〈 0. 05); but were not related to age or tumor size( all P〈 0. 05). By Pearson correlation analysis,the expression of Bcl-2 and Bax protein in LSCC patients was positively correlated with the expression of VEGF-C( r = 0. 762,P〈 0. 05; r = 0. 703,P〈 0. 05).
