摘要
目的:建立清热解毒冲洗液的质量控制方法。方法:采用薄层色谱法对方中当归、苦参、大黄、赤芍进行定性鉴别,采用高效液相色谱法测定清热解毒冲洗液中大黄有效成分大黄酸、大黄素、大黄酚的含量。结果:薄层色谱特征斑点清晰,专属性强,阴性对照无干扰;高效液相色谱法测定大黄酸在0.056~0.56μg范围内线性关系良好(r=0.999 8,n=5),平均回收率为98.47%(RSD=0.77%,n=5);大黄素在0.179~1.79μg范围内线性关系良好(r=0.999 7,n=5),平均回收率为98.58%(RSD=0.51%,n=5);大黄酚在0.035~0.35μg范围内线性关系良好(r=0.999 0,n=5),平均回收率为98.63%(RSD=0.66%,n=5)。结论:该方法简便可靠、专属性强、重复性好,可用于该制剂的质量控制。
Objective:To establish the method for quality control of heat-clearing detoxifying flush fluid.Methods:DangGui(Angelica sinensis),kuh-seng,Da Huang(Rheum palmatum L.) and Chi Shao(Radix Paeoniae Rubra)in the prescription were identified qualitatively by thin-layer chromatography(TLC),the contents of the active ingredients of Da Huang including chrysophanic acid,emodin and rheic acid were measured by HPLC.Results:TLC was featured by clear spots,strong specificity,no interference with negative controls; the results of HPLC showed that chrysophanic acid had good linear relationship in the range between 0.056~0.56 μg(r=0.999 8,n=5),average recovery rate was 98.47%(RSD=0.77%,n=5); emodin presented good linear relationship in the range between 0.179~1.79μg(r=0.999 7,n=5),average recovery rate was 98.58%(RSD=0.51%,n=5); rheic acid demonstrated good linear relationship in the range between 0.035~0.35 μg(r=0.999 0,n=5),average recovery rate was 98.63%(RSD=0.66%,n=5).Conclusion:The method,simple,convenient,reliable,specific and reproducible,could be used for quality control of the preparation.
