摘要
目的建立高效液相色谱法(HPLC)测定含精氨酸的注射用灯盏花素中精氨酸的含量。方法采用的色谱柱为Agilent TC-C_(18)(4.6mm×250mm,5μm),50mmol·L^(-1)磷酸二氢钾∶乙腈(95∶5)为流动相,检测波长206nm。结果精氨酸在200μg·mL^(-1)至1500μg·mL^(-1)之间呈良好的线性关系,相关系数(r)为0.9996,回收率为99.93%,相对标准偏差(RSD)为0.9%,n=9。用该法测定注射用灯盏花素中精氨酸的平均含量约为11.79mg。结论该法专属性好、重复性好、快速、准确,能作为部分含精氨酸的注射用灯盏花素中精氨酸含量测定的方法。
OBJECTIVE To establish a HPLC for determination of Arginine in Breviscapine for Injection. METHODS Agilent TC-C_(18)( 4. 6 mm × 250 mm,5μm) was used as chromatographic column,50 mmol·L^(-1) potassium dihydrogen phosphate ∶ acetonitrile( 95 ∶ 5) as mobile phase,and the detection wavelength was at206 nm. RESULTS The liner correlation observed was in the rang of 200μg · mL^(-1) to 1500μg · mL^(-1),r =0. 9996,The average recovery was 99. 93%,RSD was 0. 9%,n = 9. The average content of Arginine in Breviscapine for Injection was about 11. 79 mg by using this method. CONCLUSION This method is good specificity,repeatability,rapid and accurate,and can be used as part of the argrinine in Breviscapine for Injection,the method for the determination of arginine content.
