中国荷斯坦奶牛PRL基因第3外显子多态性对牛乳腺上皮细胞增殖的影响

摘要 为了研究催乳素(prolactin,PRL)基因6 216位点(A/G)突变对牛乳腺上皮细胞增殖能力的影响,试验通过体外合成PRL基因编码区(CDs区)及突变后的PRL基因CDs区PRL-mut构建真核表达载体,设置PRL转染组、PRL-mut转染组、空载转染组,通过q PCR和免疫印迹试验(Westernblot)在细胞水平上验证了PRL和PRL-mut的功能,检测了细胞的增殖水平。结果表明:荧光显微镜下三组均有较强的绿色荧光;与PRL转染组相比,PRL-mut转染组可显著增加PRLP、Stat5a和SREBP的mRNA和蛋白质表达水平(P〈0.05);与PRL转染组、空载转染组相比,PRL-mut转染组细胞增殖显著增加(P〈0.05)。说明PRL-mut基因可显著促进牛乳腺上皮细胞增殖。
