浙江省钱塘江流域抗生素抗性基因监测分析 被引量:1

Monitoring and analysis of antibiotic resistance genes in Qian Tang River basin of Zhejiang Province

摘要 目的研究钱塘江流域抗生素抗性基因分布情况。方法本研究以钱塘江流域上游、中游、下游共18份水样为研究对象,监测四环素、磺胺二甲嘧啶、氨苄青霉素的耐药菌分布,用PCR检测代表抗性基因tet A、sul I和ctx-1,用荧光定量PCR检测其Ct值和相对表达量,分析钱塘江流域耐药菌变化趋势。结果 18份水样中氨苄青霉素、磺胺二甲嘧啶、四环素的耐药菌生长阳性率分别为100.00%、88.89%和61.11%;PCR检测各水样混合微生物DNA中tet A、sul I和ctx-1,目的条带阳性率分别为70.59%、47.06%和52.94%;荧光定量PCR结果显示自上游到中游,对三类抗生素耐药性均呈上升趋势,居民区河水耐药菌表达量平均多于钱塘江水,而sul I相对表达量最高。结论钱塘江流域存在tet A、sul I和ctx-1等ARGs的污染,变化趋势与人为活动的富集污染程度、地理环境有关。 Objective To know the distribution of antibiotic resistance genes in Qian Tang River. Methods Eighteen water samples from upper and lower reaches of Qian Tang River were collected for determining the existence of tetracycline,sulfamethazine and ampicillin resistant bacteria. The antibiotic resistance gene,tet A,sul I and ctx-1,were detected by PCR and the genes' Ct value and relative expression value were examined by fluorescent quantitative PCR. Results The positive rates of ampicillin,Sulfanilamide and tetracycline resistance bacteria in 18 water samples were 100. 00%,88. 89% and 61. 11%,respectively; the positive rates of target gene tet A,sul I,and ctx-1 were 70. 59%,47. 06% and 52. 94% in the mixed microbial DNA. Fluorescence quantitative PCR results showed that the expressions of the three antibiotic resistance genes were increasing from upper stream to midstream,and the antibiotic resistance gene expression showed a rising trend in the samples of residential water from upstream to downstream and higher than Qian Tang River. The relative expression of sul I was the highest.Conclusion There are antibiotic resistance genes pollution of tet A,sul I and ctx-1 in the Qian Tang River basin,and the variation and degree of pollution are related to the enrichment of human activities and the geographical environment.
