摘要
目的:制备辛夷挥发油壳聚糖微球和优选制备工艺。方法:采用复乳-乳化交联法制备辛夷挥发油壳聚糖微球,以粒径、包封率、载药量为评价指标,通过单因素试验考察壳聚糖浓度、投药量、油水比、搅拌速度对制备工艺的影响,并设计正交试验优化制备工艺。结果:微球最佳制备工艺为:壳聚糖浓度为3%,辛夷挥发油和壳聚糖比例为1,油水体积比为4,搅拌速度为600r/min。所得微球粒径分布在30~70μm为69.37%,载药量为6.03%,包封率为28.66%。通过光学显微镜和电镜扫描,可看到微球球形圆整,分布均匀。结论:辛夷挥发油壳聚糖微球最优制备工艺稳定可行。
Objective:To prepare the chitosan microsphere of the volatile oil of Magnoliae Flos and optimize the preparation process.Methods:The chitosan microspheres of volatile oil of Magnoliae Flos were prepared by the multiple emulsion cross-linking method,setting the particle size,drug loading,encapsulation efficiency as evaluation indexes,the effects of chitosan concentration,dosage,oil-water ratio and stirring speed on the preparation process were investigated by single factor test,and the orthogonal experiment was also designed to optimize the preparation process.Results:The optimal preparation process of microspheres was as follows:the concentration of chitosan was 3%,the feed ratio of the volatile oil of magnoliae flos and chitosan was 1,the ratio of oil to water was 4,and the stirring speed was 600 r/min,and.The particle size distribution of the microspheres was 69.37%,the drug loading was 6.03%,and the encapsulation efficiency was 28.66%.Through the optical microscope and the scanning electron microscope,the microspheres were regular round in its morphology and evenly distributed.Conclusion:The optimum preparation process of the volatile oil of Magnoliae Flos chitosan microspheres is stable and feasible.
