猪鼻支原体的分子鉴定及进化分析

摘要 本研究对青藏高原地区青海猪鼻支原体分离株(Mhr-QH1)进行了Mhr-p37基因的克隆鉴定及测序分析,并利用Mhr-16S rRNA基因与Gen Bank中相关菌株基因进行了基因同源性比较和遗传进化分析。Mhr-p37基因和16S基因PCR扩增测序结果显示,获得核苷酸序列长度分别为346bp和1500bp,Mhr-QH1-p37基因核苷酸序列与中国株、法国株和美国株的同源性达到100%;同样16S rRNA基因与巴西分离株的16S rRNA基因同源性也为100%,与日本、瑞典和美国分离株的同源性为99%。进化树分析发现:Mhr-QH1分离株与巴西株聚为组成一个微小分支,与美国株和巴西株共聚为同一个大支上,并与其他Mycoplasma spp.种系进化距离较远。因此,本研究对青海猪鼻支原体菌株的Mhr-p37基因序列和蛋白氨基酸序列分析,及16S rRNA基因系统进化研究的结果,有望为我国猪鼻支原体病的分子流行病学调查提供一定的数据参考,同时提示猪场搞好饲养管理是预防本病的关键。 In the present study,the Mhr-p37 gene was cloned,sequenced and molecular identificated of the Mycoplasma hyorhinis QH isolate from Qinghai-Tibet Plateau,and the gene homology and genetic evolution analysis of16 S rRNA gene was performed with that of related genes in Gen Bank. The PCR amplification and sequencing results of Mhr-p37 and 16 S rRNA genes showed that the obtained nucleotide sequences were 346 bp and 1500 bp in length,respectively. The nucleotide sequence of Mhr-QH1-p37 gene was with 100% homology to that of China,French and American strains. Similarly,the 16 S rRNA gene was also with 100% homology to that of Brazilian isolate,and with99% homology to which of Japan,Sweden and the United States strains. Phylogenetic analysis revealed that the Mhr-QH1 isolate and the Brazilian strain were clustered to form a tiny branch,and which was co-polymerized with the American and Brazilian strains on the same branch,and which was evolved further away from other Mycoplasma spp..Therefore,the analysis of the Mhr-p37 gene and amino acid sequences of the M. hyorhinis strain and the result of the16 S rRNA gene phylogenetic analysis,which are expected to provide data references for the molecular epidemiological investigation of M. hyopsiosis in China,and prompte good feeding and management for preventing the disease in the pig farms.
