摘要
目的探讨超声介导载miR-34a脂质微泡对小鼠U14宫颈癌皮下移植瘤的抑制作用。方法(1)制备载miR-34a阳离子脂质微泡,检测其一般特性;(2)建立小鼠U14宫颈癌皮下移植瘤模型,随机分组为对照组、空阳离子脂质微泡组、载miR-34a阳离子脂质微泡组;(3)根据分组给予微泡+超声辐照的处理,绘制各组肿瘤生长曲线,计算抑瘤率,实时荧光定量PCR技术检测miR-34a、c-met、bax基因表达情况,免疫组织化学法检测c-Met、Bax蛋白表达情况。结果(1)微泡镜下呈球形,平均粒径(1.2±0.24)μm。(2)载miR-34a阳离子脂质微泡组小鼠肿瘤生长较其他小组缓慢,肿瘤体积明显较小(P<0.05),Bax表达上调,c-Met表达下调(P<0.05)。结论超声介导载miR-34a阳离子脂质微泡可明显抑制小鼠U14宫颈癌皮下移植瘤生长,促进肿瘤细胞凋亡。
Objective To investigate the inhibitory effect of ultrasound-mediated miR-34a-loaded lipid mi- crobubbles on U14 cervical cancer transplantation tumor in mice. Methods The miR-34a-loaded cationic lipid microbubbles was prepared and its general characteristics were detected. The mice model of U14 cervical cancer transplantation tumor was established. The mice were rando mLy divided into three groups : control group, empty cationic lipid microbubbles group, miR-34a-loaded cationic lipid microbubbles group. After than, the mice were treated with microbubbles and ultrasonic radiation according to different groups, the tumor growth cm-ve was drew according to the tumor volume, and the rate of tumor inhibition was calculated after sacrificing the animals at the end of experiment. The expressions of miR-34a, e-met and bax were detected by real-time fluorescence quantitative PCR, and the expressions of c-Met and Bax protein were detected by immunohistochemistl7. Results ( 1 ) The mi- crobubbles were round and uniform with the mean size of 1.2μm. 2. The tumor growth was slower in miR-34a- loaded cationic lipid microbubbles group than in other groups. The tumor volume was significantly decreased (P 〈 0.05), the expression of Bax was up-regulated and the expression of c-Met was down-regulated (P 〈 0.05). Conclusions Ultrasound- mediated miR- 34a- loaded cationic lipid microbubbles can significantly inhibit the growth of U14 cervical cancer transplantation tumor and promote tumor cell apoptosis.
