消渴通痹颗粒对体外高糖环境下EPCs迁移及管腔形成的影响

摘要 目的探究益气活血中药消渴通痹颗粒对体外高糖环境下内皮祖细胞(endothelial progenitor cells,EPCs)迁移及管腔形成的影响。方法选取24只清洁级SD大鼠,随机分为空白组(6只)与给药组(18只)。给药组随机均分为3组,分别按消渴通痹颗粒大、中、小剂量给药设为大、中、小剂量组,空白组饲喂等量生理盐水,连续给药7 d后采血并离心制备血清。空白组血清随机均分为2组,一组是加入伊红美蓝培养基-2(eosin-methylene blue medium-2,EBM-2)完全培养液为对照组;另一组是加入30 mmol/L葡萄糖的EMB-2完全培养液为高糖对照组。给药组血清均分别加入30 mmol/L葡萄糖的EMB-2完全培养液;抽取小鼠骨髓,从中分离EPCs,进一步制备细胞悬液,培养后用CD34抗体及DAPI进行染色鉴定并观察不同组培养液中其迁移能力及管腔形成能力。结果高糖对照组EPCs迁移数低于对照组,差异具有统计学意义(P<0.05),大、中、小剂量组EPCs迁移数高于高糖对照组,差异具有统计学意义(P<0.05);大、中、小剂量组管状形成百分比均高于高糖对照组,差异具有统计学意义(P<0.05)。结论高糖环境下,消渴通痹颗粒能显著改善EPCs迁移的能力与迁移数,改善EPCs管腔形成的能力。 ObjectiveTo investigate the effects of qi-reinforcing and blood circulation-promoting Xiaoketongbi granules on the migration and angiogenic ability of EPCs under high glucose. MethodsIn this study, 24 clean SD rats were randomly divided into the blank group(n=6)and administration group(n=18). Then, the administration group was randomly divided into the high dose group, medium dose group and small dose group according to the dosage of Xiaoketongbi granules. The blank group was fed with normal saline of the same amount. Serum was collected after 7 days of continuous administration via blood sampling and centrifugation. The serum of the blank group was randomly divided into two groups: one control group in which the complete medium of eosin-methylene blue medium-2(EBM-2)was added, and one high glucose control group in which the complete medium of EMB-2 with 30 mmol/L glucose was used. The complete medium of EMB-2 with 30mmol/L glucose was added to the serum of the administration groups. EPCs were isolated from the bone marrow of mice to prepare cell suspension. After culture, the cells were stained with CD34 antibody and DAPI, and their migration ability and angiogenic ability in different culture media were observed. ResultsThe number of EPCs on migration in the high glucose control group was smaller than that in the control group, and the difference was significant(P〈0.05). The numbers of migratory EPCs in high, medium and small dose groups were larger than those in the high glucose control group(P〈0.05). The percentage of tubular formation in high, medium and small dose groups was significantly higher than that in the high glucose control group(P〈0.05). ConclusionXiaoketongbi granules can significantly improve the migration and angiogenic ability of EPCs under high glucose.
