摘要
为建立微波消解-电感耦合等离子体质谱(ICP-MS)法测定药用植物白术根茎中微量元素的含量,本研究采用HNO3-H2O2体系,将3批湖南产白术根茎粉碎(过200目筛),通过湿法消解后,以In、Rh为内标元素,用ICP-MS法测定白术根茎中48种微量元素的含量。结果表明,测定的各元素线性关系良好,各标准曲线相关系数r≥0.999 6;检测限为0.000 1 g/L^0.160 0 g/L;精密度、稳定性和重复性RSD分别小于4.7%、4.9%和4.4%;加样回收率为96.9%~101.7%,RSD 0.3%~3.5%;其中B、V、Cr、Mn、Co、Ni、Cu、Zn、Se、Rb、Sr、Mo、Sn等人体必需微量元素含量较高,Pb、As、Cd、Hg等重金属元素含量均低于《中国药典》2015版对重金属元素的限度要求。该方法灵敏度高、精密度高、线性范围广、准确性好,可快速对白术根茎中的48种微量元素同时进行测定,填补了药用植物白术根茎中微量元素研究的空白,为今后深入研究白术中各元素存在的形态,为进一步开展其质量控制、活性筛选、安全性评价等研究提供科学依据。
To establish an inductively coupled plasma spectrometry(ICP-MS) with microwave digestion method for the determination of 48 kinds of trace elements inAtractylodes macrocephala Koidz., fill the blank of studying on the trace elements of this plant, and provide scientific basis for its quality control and further research.Three batches ofA. macrocephala Koidz. produced in Hunan Province were used as samples. The contents of trace elements were determined by inductively coupled plasma mass spectrometry (ICP-MS) after the samples were diluted with wet digested in HNO3-H202 system, then with In, Rh as the internal standard.The 48 trace elements showed good linearity in the selected concentration ranges, with the correlation coefficients of the standard curve were over 0.999 6. The detection limits of the 48 elements were in the range of 0.000 1 ~ag/L-0.160 0lag/L.The RSD values of precision, stability, and repeatability were less than 4.7%, 4.9% and 4.4% respectively, which all met the demands of quantitative analysis. The recovery was 96.9%- 101.7%, while the RSD was 0.3%-3.5%. The contents of the trace elements B, V, Cr, Mn, Co, Ni, Cu, Zn, Se, Rb, Sr, Mo, Sn which were essential to hu- man were high.The contents of the heavy metal elements including Pb, As, Cd, Hg were under the limit requirements of Chinese Pharma- copoeia (2015).The method has high sensitivity, high precision, wide linear range and good accuracy, which can be used to determine the contents of 48 trace elements in the root ofA. macrocephala Koidz. A. macrocephala Koidz. has rich contents of Cu, Sr, Mn, Zn, Sn, B, Cr, Rb, Ni and other trace elements essential to human. These elements have direct or indirect effects on the efficacy of this plant when it participate in vivoprocess.The analysis of trace elements can provide certain references for the elementary speciation, quality control, activity screening, safety evaluation, and clinical application ofA. macrocephala Koidz..
