摘要
目的建立快速灵敏的高效液相色谱法测定大鼠血浆中灯盏花乙素含量的方法。方法以原儿茶醛为内标,乙酸乙酯作为萃取试剂进行液-液萃取。采用Kromasil C18色谱柱(5μm,4.6 mm×250 mm),以乙腈-0.02 mg/L磷酸氢二钾(15∶85)为流动相进行洗脱,流速为1.0 m L/min,进样量为20μL。结果灯盏花乙素在0.1~100μg/L范围内线性关系良好;灯盏花乙素的提取回收率大于80%;方法的日内及日间精密度RSD均小于5.74%。准确度在96.24%~97.89%之间。结论该方法操作简便、快捷、灵敏度高,适用于灯盏花乙素在大鼠体内的药代动力学研究。
Objective The study established HPLC method for the determination of Breviscapine from the Chinese herb Erigeron breviscapinus. MethodsThe experiment using protocatechualdehyde as an internal standard(IS) in rat plasma. Plasma sample pretreatment involved a one-step liquid-liquid extraction with ethyl acetate. Chromatographic separation was performed on a Kromasil C 18 column(4.6 mm × 250 mm, 5 μm) with isocratic elution using acetonitrile-0.02 mg/L dipotassium phosphate(15∶85) as mobile phase at a flow rate of 1 m L/min. Results The breviscapine had good linearity in the range of 0.1 μg/m L to 100μg/m L. The extraction recoveries of breviscapine were more than 80%. The intra-and inter-day precision values of analysis methodwere not more than 5.74%, and accuracy was from 96.24% to 97.89%. Conclusion The method is simple, rapid and sensitive, which is suitable for pharmacokinetics study of the breviscapine in rats.
