大鼠及人先天性肛门直肠畸形中Wnt5a及BMP7基因的表达研究 被引量:2

摘要 目的探讨Wnt5a及骨形成蛋白7(BMP7)基因在先天性肛门直肠畸形中的表达。方法将已受孕的大鼠(35只)根据是否胃管注入乙烯硫脲(ETU)以及是否存在肛门直肠畸形分为对照组(12只)、ETU无畸形组(8只)和ETU畸形组(15只)。RT-PCR检测妊娠第13、14、15、16天大鼠直肠及泄殖腔Wnt5a和BMP7的mRNA表达;Western blot检测妊娠第16天大鼠直肠Wnt5a和BMP7的蛋白表达;高位畸形、低位畸性患者及非胃肠道疾病死亡,分为高位畸形组、低位畸形组和对照组,RT-PCR和Western blot分别检测直肠末端Wnt5a和BMP7的mRNA和蛋白表达。结果 ETU组大鼠的Wnt5a及BMP7基因的mRNA表达在4个时间点均显著低于对照组,差异均有显著统计学意义(P<0.01);ETU畸形组大鼠的Wnt5a和BMP7的蛋白表达显著低于对照组,差异均有显著统计学意义(P<0.01),而大鼠ETU无畸形组Wnt5a和BMP7的蛋白表达与对照组比较差异均无统计学意义(P>0.05);人高位畸形组大鼠的Wnt5a和BMP7的mRNA和蛋白表达均显著低于对照组,差异均有显著统计学意义(P<0.01),而人低位畸形组Wnt5a和BMP7的mRNA和蛋白表达与对照组比较差异均无统计学意义(P>0.05)。结论 Wnt5a和BMP7基因的表达下降可能与先天性肛门直肠畸形有关。 Objective To investigate the expression of Wnt5 a and BMP7 genes in congenital anorectal malformations. Methods The pregnant rats( 35 rats) were divided into the control group( 12 rats),the ETU free group( 8 rats) and the ETU malformed group( 15 rats) according to whether injected with ETU and if there was anorectal malformation. The expression of Wnt5 a and BMP7 mRNA were detected on 13 d,14 d,15 d,and 16 d of gestation in rectum and cloaca of rats by RT-PCR. The expression of Wnt5 a and BMP7 protein on sixteenth day of pregnancy rats rectum were detected by Western blot. Patients with high malformation,low malformation,and non-gastrointestinal diseases were divided into high malformation group,low malformation group,and control group. RT-PCR and Western blot were used to detect the mRNA and protein expression of BMP7 and Wnt5 a. Results Wnt5 a and BMP7 gene mRNA expression in rat ETU group in the 4 time points were significantly lower than the control group( P〈0. 01). The expression of Wnt5 a and BMP7 protein in rat ETU abnormal group was significantly lower than the control group( P〈0. 01).The expression of Wnt5 a and BMP7 protein in rat ETU without abnormal and the control group had no significant difference( P〈0. 05). The mRNA and protein expression of Wnt5 a and BMP7 in human high deformity group were significantly lower than those of the control group( P〈0. 01). The mRNA and protein expression of Wnt5 a and BMP7 in human low malformation group and the control group had no significant difference( P〈0. 05).Conclusion The decreased expression of Wnt5 a and BMP7 genes may be associated with congenital anorectal malformations.
