摘要
目的观察自拟复方中药汤剂治疗大鼠肝纤维化时肝组织中基质金属蛋白酶-2(MMP-2)、金属蛋白酶组织抑制因子-2(TIMP-2)、结缔组织生长因子(CTGF)的表达,初步探讨自拟复方中药汤剂治疗肝纤维化的机制。方法 100只雄性SD大鼠建立四氯化碳大鼠肝纤维化模型,造模后分别以含低、中、高剂量姜黄的自拟复方中药汤剂给随机分组的大鼠灌胃处理,以小柴胡汤和复方鳖甲软肝片为阳性对照,12周后处死大鼠,留取同一部位新鲜肝组织,行HE染色,光镜下观察病理学改变,并按SSS计分系统进行评分,免疫组织化学染色检测肝组织MMP-2、TIMP-2、CTGF表达,专业图像分析软件(Axio Vision 4.5版,Germany)进行图像分析。结果与模型对照组比较,各治疗组肝纤维化评分明显降低(P<0.05),而MMP-2的表达量增加(P=0.001),TIMP-2的表达量减少(P<0.05),CTGF阳性表达明显减少(P<0.05)。结论自拟复方中药汤剂可以减轻肝纤维化大鼠的肝纤维化病理改变;可能与其抑制促肝纤维因子CTGF、TIMP-2的表达,促进抗肝纤维化因子MMP-2表达增多有关。
Objective To observe the changes of expression of matrix metallo Proteinases-2(MMP-2),tissue inhibitor of metalloproteinases-2(TIMP-2) and connective tissue growth factor(CTGF) in fibrotic liver tissue of rats treated with the compound traditional Chinese medicine decoction and to study the mechanism of them. Methods Hepatic fibrosis model was established in 100 male SD rats, all rat models were established by intraperitoneally injection of carbon tetrachloride. The fibrotic groups were treated with compound traditional Chinese medicine decoctions containing low, middle and high doses of curcumin by intragastric administration to the fibrotic rats respectively, treatments with Xiaochaihu decoction and compound Biejiaruangan tablets were made as positive controls. Then all the rats were killed after 12 weeks. HE staining were conducted in fresh hepatic tissues from the same area to observe pathological variations. Grades of hepatic fibrosis were evaluated according to the SSS system. The expressions of MMP-2, TIMP-2 and CTGF in liver tissues were examed by immunohistochemical staining and professional software for image analysis was used. Results Compared with the model control group, the score of hepatic fibrosis of each treatment group decreased significantly(P〈0.05), the expression of MMP-2 increased(P=0.001), the expression of TIMP-2 decreased(P〈0.05), and the expression of CTGF decreased significantly(P〈0.05). Conclusion The compound traditional Chinese medicine decoction can alleviate the pathological changes of hepatic fibrosis in rats, which may be related to the inhibition of the expression of CTGF and TIMP-2 and the increase of the expression of anti-fibrosis factor MMP-2.
