摘要
目的:探讨自噬相关蛋白Beclin1及微管相关蛋白轻链3-Ⅱ(LC3-Ⅱ)在风湿性瓣膜病组织中的表达及意义。方法:选取2015年10月至2017年10月广西医科大学第一附属医院收治的风湿性心瓣膜病主动脉瓣狭窄或风湿性心脏病联合瓣膜病变主动脉瓣狭窄病例为实验组,同期选取A型主动脉夹层需行Bentall手术或Bentall+Suns手术健康主动脉瓣病例为对照组;主动脉根部切开,灌注心脏停搏液,心脏停跳后即刻切取主动脉瓣;采用免疫组化染色及western blotting检测瓣膜组织中Beclin1和LC3-Ⅱ蛋白的定位及表达差异。结果:实验组的主动脉瓣膜组织中Beclin1和LC3-Ⅱ蛋白表达含量高于对照组(P<0.05)。结论:风湿性瓣膜病存在自噬现象,自噬因素可能参与风湿性瓣膜病的发病。
Objective:To investigate the expression of autophagy associated protein Beclin1 and microtubuleassociated protein light 3-Ⅱ(LC3-Ⅱ)in rheumatic valve disease.Methods:The patients with rheumatic aortic stenosis or rheumatoid joint valve lesions aortic stenosis treated in our hospital from October 2015 to October 2017 were selected as an experimental group.The patients with type A aortic dissection treated with Bentall surgery or Bentall plus Suns surgical healthy aortic valve were defined as a control group.Aortic valve was removed immediately after cardiac arrest.The protein expressions of Beclin1 and LC3-Ⅱin the valve tissues were determined by immunohistochemistry and western blotting,respectively.Results:The expressions of Beclin1 and LC3-Ⅱin aortic valve tissues in the experimental group were higher than those in the control group(P〈0.05).Conclusion:Autophagy exists in rheumatic valvular disease,and autophagy factors may be involved in the pathogenesis of rheumatic valvular disease.
